Nerve growth factor (NGF) is a kind of very important peptide to the human body. It can strongly affect cell division and growth and it can cure nerve system disease. But NGF is easy to lose biological activity quickly in water solution and is sensitive to many conditions such as pH, high temperature, and so on .The use of NGF is limited, as it can't cross the BBB freely. So this experiment is trying to change dosage form in order to solve the problem of stability and the penetration of the BBB.An analytic method was developed to determinate the NGF with Brilliant yellow by the Rayleigh light scattering method using Triton X-100 as sensitizer. NGF liposome was obtained by the sonication method. Through ultracentrifuge method the encapsulation efficiency (EE) was measured easily. Single factor investigation was carried out , and optimal formulation was decided . With the optimal formulation, the EE of the liposome was about 34%. Use the 0.22μm micropore membrane for sterilization which did not affect the EE.The physical properties of liposome were described. It was shown that the liposome carried positive charge. The laser scattering measurement showed that NGF particle was nanoparricle with mean diameter <50 nm. The flexibility test showed that the liposome studied in this paper had more flexibility than normal liposome.The safety experiments of liposome such as vessel irritation and hemolysis were tested.
