Methods: The RT-PCR technique was applied for the assay of the expression of wild type and mutant P53 gene in MDA-MB-231 cells and MCF-7 cells. Results: The expression of mutant P53 gene was detected and increased by Emetine treatment (P<0.05).The expression of wild type P53 gene was not found in control and treat groups of MDA-MB-231 cells. We detected the wild type P53 was expressed in MCF-7 cells, and don't have great change after Emetine treatment (P>0.05). The mutant P53 were not found in control and treat groups of MCF-7 cells. Conclusion: The expression of mutant P53 was increased by Emetine treatment as compared with control groups in MDA-MB-231 cells, while the expression of wild type P53 was not great difference by Emetine treatment as compared with control groups in MCF-7 cells. The influences of Emetine on gene expression of mutant P53 than wild type P53 are relative selectivity. | Keywords/Search Tags: | breast cancer, P53, NMD, emetine, human breast cancer cells, Emetine, Y14, UPF1 | PDF Full Text Request | Related items |
