Objective: To investigate G. Pentaphyllum' s influence on the activity of Glutathione peroxidase (GHS-PX) and Catalase (CAT) in the skin of hireless photoageing mice as well as the content of hydrogen peroxide (H2O2) in their skin, and to investigate G. Pentaphyllum' s effect on the structure of the hireless photoageing mice.Methods: Healthy KM hireless mice were divided into five groups randomly and gavaged respectively, that is negative Control Group (A), Model Group (B), Low- G. Pentaphyllum -Dosage Group (C), Medium-G. Pentaphyllum -Dosage Group (D), High- G. Pentaphyllum -Dosage Group (E). The mice in B, C, Dand E group were irradiated to simulate skin photoageing by long-term ultraviolet irradiation, at the same time, group B were given vitamin E while those in C, D, and E were given G. Pentaphyllum. Staining skin with HE, Under 40 times light microscope, the structure of dermal was observed. While the changes of the content of H2O2, as well as the activity of GHS-PX and CAT in the skin were measured by biochemical test method in each group of the experiment.Results: In model group, the activity of GHS-PX and CAT were decreased , while the content of the H2O2 was increased seriously (P <0. 01) .The skin biopsies appear photoageing look. Compared with the group B, the activity of CAT in group D was made a great progress (P <0. 01) ; as well as the activity of GHS-PX was increased and the content of H2O2 was reduced in dermal in group C and E, furthermore the latter is dependent on dosage obviously (P<0. 01, P<0. 05) .Conclusions: G. Pentaphyllum has the protective effect on the skin of the photoageing hairless mice. The mechanism is that on one hand, G. Pentaphyllum could eliminate free radicals itself; on the other hand, by increasing the activity of GHS-PX and CAT, it could inhibit the oxidative damages of ultraviolet and skin photoaging.
