| Objective: Through the observation of the apoptosis of lymphocyte in the spleen of ITP model mice and the expression of bcl-2 apoptosis protein, discuss the effect of abnormal apoptosis of immune cells on the disease, and analyze the effect and mechanism of purpura granule core formula from the aspects such as peripheral platelet count and immune cell apoptosis index.Method: Establish ITP model by applying immune method; namely, establish the model through injecting exogenous APS. Applying Blood Cell Counter to examining peripheral platelet count. Applying Flow Cytometry to observing the apoptosis of lymphocyte in the spleen of ITP model mice and the expression of bcl-2 apoptosis protein. Results: Comparing to model group, the peripheral platelet count of purpura granule core formula group increases obviously (P<0.01), the comparison between Prednisone group and Xuemeian group has no statistical meaning. The apoptosis rate of model lymphocyte is obviously lower than normal group, the comparison between the two groups has statistical meaning (P<0. 05). The lymphocyte apoptosis rate of purpura granule core formula group and Prednisone group is obviously higher than model group (P<0. 05), the comparison between groups has no statistical meaning. The expression of bc L-2 apoptosis protein of model group is obviously higher than normal group (P<0. 05). The bcl-2 apoptosis protein rate of purpura granule core formula group and Prednisone group is lower than model group, and the comparison between groups has no statistical meaning.Conclusion: Purpura granule core formula can apparently improve the peripheral platelet count of ITP mice and effectively control the bleeding. Purpura granule core formula can restrain apoptosis of lymphocyte and enhance apoptosis expression of ITP mice spleen lymphocyte by reducing expression rate of bc L-2 apoptosis protein of ITP mice spleen lymphocyte and exerts the function of treating ITP through regulating the apoptosis function of immune cells. |
