It has been confirmed that the traditional Chinese medicine can treat DN by activating blood circulation to dissipate blood stasis. Xiao Tang Capsule (XTC) was the compound preparation of our department advanced development, and it has got satisfaction effect in the clinic. In the experiments, the model of DM was established by njecting alloxan and nephrectomy in rats. The normal situations, the quality of the bodies and kidneys, fasting blood glucose, urine small protein, renal function, angiotensin II and aldosterone were observed. The protective effect of XTC on the rat kidney of DM and the initial mechanism were researched for proving the dependable experimental evidence to treat DN in the future.Experiment I Establishment of the diabetic model in ratsOBJECTIVE: To investigate the optimal dose of alloxan (ALX) and the time of establishment the diabetic model in rats. METHODS: The experiment rats were divided into five groups: normal group, cutting left kidney group, model I group, model II group and model III group. The diabetic model was established by injecting alloxan and nephrectomy in model I , II and model III groups. The normal situations were observed, while blood and urine glucose were checked and the renal pathological change were observed. RESULTS: Comparing with the normal group, the rats were tired, debilitational, urorrhagia, food-intake increasing, more hydroposia and body weight loss in the model groups, while they were fond of eating sweet food. Blood glucose were between 5.2~6.0mmol/L in normal group and cutting left kidney group. After The diabetic model was established, at the 4th day , the blood glucose were higher than 16.7mmol/L in the model groups, and the blood glucose of four rats were higher than 25mmol/L in the model III group.
