Musk The Yellow Xiaoliu Side, Against Hepatocellular Carcinoma Cell Invasion And Metastasis Of Experimental Research

Objective: To study the inhibitory effect of the rabbit serum containing SheHuangXiaoLiu Fang(SHF) on invasive and adhesive ability of SMMC-7721 cells,and to research into the possible mechanisms.Methods: SMMC-7721 hepotocellular carcinoma cells were exposed to the rabbit serum containing SHF,excluding SHF respectively .Cell number and inhibitory rate were evaluated every 24 hours.Effect of SHF on SMMC-7721 cell membrane intercellular adhesive molecule-1(ICAM-1) expression was observed by immunofluorescence flow cytometry;effect on invasive ability of SMMC-7721 cells and the detachment of which attached to fibronectin(FN) by cell migration experiment.Effect of SHF on nm23-hl expression was assessed by immunohistochemistry;effect on adhesion of SMMC-7721 cells with FN was observed by MTT method.Results: The growth of SMMC-7721 cells incubated with SHF was inhibited conspicuously ,and the top inhibitory rate of tumor cells was reaching to 66.25% in the 5^th day.ICAM-1 expression in SMMC-7721 cells incubated with SHF was significantly lower than that in cells did not treat with SHF,and the tumor cells treated with SHF could significantly increase the expression of nm23-h1.SHF could inhibit the invasive ability of SMMC-7721 cells,and it could also inhibit the adhesion of tumor cells attaching to FN, even made the adhesive cells exfoliated.Conclusion: SHF have the effects inhibiting the adhesion and invasion of SMMC-7721 cell.It can enhance the expression of nm23-hl and reduce that of ICAM-l,which might be the molecular machanism .
