Chai Huangyi Kidney Side Kidney Damage Mechanism Of Their Underlying Mechanisms

Objective: To investigate the therapeutic and diuretic effects of CHC on experimental rats with nephropathy and overhydration. To investigate mechanism of CHC on nephropathy induced by PAN.Methods:The model of glomerular and interstitial nephritis induced by puromycin aminonucleoside(PAN) and the model of chronic renal insufficiciency induced by classical method(removal of the right kidney and ligation of 2/3 the left kidney artery)were applied to observe the effect of CHC on the amounts of 24h urine protein excretion and parameters of blood biochemistry in rats. Meanwhile, the renal tissue obtained was processed for light microscopy and compared the degree of renal tissue injury by semiquantitative scoring system. Immunohistochemistry staining and quantitative assay by computer system were used to investigate the pathological changes of kidney and expression of α -smooth muscular action ( α -SMA) and transforming growth factor β 1(TGF-β1) of the kidney. The model of water-loaded was applied to observe the effect of CHC on urinary amount among the observed points and total urinary amount.Results: ①CHC can significantly suppress large quantity of urinary protein ,increase the level of serum proteins and serum albumin, decrease the level of cholesterol, triglyceridemia, blood urea nitrogen and creatinine, reduce the glomerular and tubulointerstitial injury index(P<0.05, P<0.01, p<0.001), as compared with model group(PAN or CRI). ②CHC can significantly raise urinary amount in the 3rd, 5th ,7th hour and total urinary amount (P<0.05, P<0.01, P<0.001), as compared with water-loaded model group. ③It showed that CHC can alleviate the glomerulosclerosis and tubulointerstitial damages, reduce the levels of expression of TGF- β1 (P<0.05) and α -SMA(P<0.001) significantly, as compared to PAN-model group.Conclusion: All these results suggest that CHC have the obvious effects on lessening protein in the urine, low blood protein levels, high cholesterol levels and swelling in experimental rats with nephropathy, improving renal function, postponing focal glomerulars clerosis and tubulointerstitial fibrosis, morethan, CHYS Compoud can obviously relieve renal fibrosis and its mechanism is related to their effects of inhibiting the expression of TGF- β1 and α -SMA...
