Objective: To observe the influence of the total polysaccharide from Chinese dodderseed (TPCD) on MSCs differentiation and the healing function of stem cells which are in the lesion skin. To explore the differences between the actions of TPCD in vitro and in vivo , and the relationship of lung and kidney.Methods: Cell Test: Choose the marrow from tibia and femur of SD rats as test objectsand do vitro studies. We add TPCD and DMSO into the different test groups to observe the results through invert microscope and apply Transcription-Polymerase Chain Reaction techniques and Immunocytochemistry techniques into this test.Lesion skin test: We cut off skin from scapular area of SD rats with the size of 1.5 × 1.5cm~2 and use the powder of total polysaccharide from Chinese dodder seed, Vaseline gauze and physiological saline gauze to treat the lesion part. Naked eye general observe the healing of the lesion skin. Kill the rats in the end of 4th week and 8th week and take the lesion skin to do tissue pathology test.Results: The shape of the mesenchyma stem cells change after the inducement of TPCD and a part of them even have the axon .The RT-PCR result shows that all the three samples have mRNA expression of nerve cell NSE. The expression of the group of TPCD is obviously stronger than the contract group and the DMSO expresses the strongest. All the three groups have the expression of GFAP and the group of TPCD stronger than contract group while the DMSO group is less strong than the contract group. Immunocytochemistry test shows that TPCD group, DMSO group and contract group have NSE expression 27.45%,41.5%,8%, and the GFAP expression 11.7%,4.323%, 5.9%.TO general observe the healing of the lesion skin, discovers the scar of the TPCD group more narrow than other groups and the growth of sparse hairs in lesion skin of TPCD group. Histopathology test shows that TPCD group has obvious differences from the other two groups in the healing of hair follicles and the blood vessels in the 4th week and after 8 weeks' treatment, and the recovery of cutaneous appendage (hair follicles, sebaceous gland) and capillary vessels differs from the other two groups, but the...
