As a plant microcomponent, Resveratrol is a polyphenolic compound produced by several species and found especially in Hu Zhang and grape and therefore can be present in human diet or beverage (red wine, for instance). There are two methods to produce Resveratrol, natural plant extract and chemical synthesis. However, They all have some disadvantages. So our study concentrated on studying the fermentation conditions of Recombinant yeast which can produce Resveratrol. Meanwhile its anti-obesity effect and preliminary mechanism were studied.Firstly, the fermentation conditions of Saccharomyces. cerevisiae FY23 which can produce Resveratrol was optimized. To detect the Resveratrol, HPLC method was set up.Then The fermentation conditions were designed as follows: culture medium------YPDmedium, which contains 1.4% glucose as carbonic source. While the cultivationconditions------seed age 36h, cultivation pH5.0, 30℃, 200 rpm/min, 72h. The content ofResveratrol reached 1.913ug/L.To detect the anti-obesity effect of Resveratrol, the diet-induced obesity (DIO) mice model was established by high fat food. Then Resveratrol was administrated orally in water. After 18 weeks, the body weight, fat content, serum cholesterol and LDL, number of fat cells per unit visual field were selected to observe the anti-obesity effect of DIO mice. Results: Resveratrol could lower the body weight and fat content about 5 gram, decrease the diameter of fat cell, lower the level of serum cholesterol and LDL, while the insulin sensitivity was improved a lot. Conclusion: Resveratrol has anti-obesity effect in diet-induced obesity.Finally, the preliminary mechanism was discussed. Energy balance exists when energy intake and expenditure are steady. That is to say, fat accumulation is because of energy imbalance. So in the experiments energy intake which equals to dietary energy intake and energy expenditure or thermogenesis were detected.
