Abstract/Summary:
Objective:⑴Observe the change of RGCs (retinal ganglions)density, Through Retrogradely labeled RGCs by fluorogold (FG) were counted in retina under the different injured time and different periods after optic nerve partially injuried in SD rats.⑵To study whether it has the ability to protect RGCs or not by inject CNTF(ciliary neurotrophic factor) into cerebrospinal fluid(CSF) after optic nerve partially injuried in SD rats.Methods:⑴Established the quantitive and various extent incomplete injured model of adult rats optic nerve by across action forceps, through Retrogradely labeled RGCs by FG were counted in retina, Under three different wounding time, 5 seconds,10seconds and 20 seconds, individually observe the change of RGCs density in 7, 14 ,21days .⑵Established crushed 5 seconds injured model of optic nerve, it was divided into 3 Groups: CNTF treating groups; rats were infected CNTF100μg or 150μg into the CSF, control group(were injected distilled water) , individually obtain retina after 7 and 14 days, through Retrogradely labeled RGCs by FG were counted in retina, Observe the density of RGCs between different groups and whether it has the difference; optic nerve was observed by light microscope and electron microscope.⑶SPSS11.0 software package served as statistical analyze tool.Results:⑴Between the groups of different wounding time, change of RGCs density is very prominent (P<0.01 ); During different periods after damaging, change of RGCs density is very prominent (P<0.01 ) between every experimental group; The relationship between the RGCs density of surviving and wounding time is very prominent (P<0.01 ).⑵7 and 14 day after damage, the difference of the availability between CNTF treating groups and control group is very prominent, RGCs density of CNTF treating groups higher obviously than control treating group, but RGCs density change does not have significance difference (P> 0.05 ) between two CNTF treating groups; ratios of totle axons to the optic nerve section area of CNTF treating groups more than control group significantly; by electron microscope , CNTF treating groups showed a significantly lower lesion of optic nerve than control treating groups.Conclusions:⑴The density of surviving RGCs was related with optic nerve wounding time and periods after damaged .⑵Offer CNTF in the CSF could provide neuroprotection for RGCs after optic nerve partial injured and showed the best effect in the 7th day , but what is the best dosage , it still needs we continue to study.
