[Objective] Cognition function of hunman is involved in learning ability , memory , attention intelligence ,and etc.The long-term repeted seizure in epilepsy usually leads to cognitive impairment ,which shows the symptoms such as memory damage intelligence breakdown ,attention disorder, and etc .The recent clinical research reveals that both the types of epilepsy and the anti-epilepsy medicines are contributing to the cognitive impairment in epilepsy patients ,the level of which has direct ratio to the seizure frequency and medicine doses .This experiment is aimed at evaluating the impairment effects of the two factors so that the doctors and the patients can avoid the adverse cause of cognitive impairment in the treatment.The Chinese medicine complex prescription has distinct advantages in the therapy of epilepsy .Caoguozhimutang is the classical and effective prescription ,according to the theory of the spleen and stomach in charge of psychomotility ,coming from Wenbingtiaobian ,used by Pro.Wang Hongtu ,to treat epilepsy .Through copious clinical and experimental research ,we find that this prescription has desirable effects in ameliorating cognitive impairment .This experiment is devised to probe the mechanism that Caoguozhimutang block the formation of cognitive impairment. [Metheds] We used the epilepsy model rats kindled by pentylenetrazol ,made them repeted seizure or lavage overdose phenytoin sodium ,and evaluated their cognitive function with Morris labyrinth.Then we test the cAMP response element-binding protein and c-fos protein variance by immunohistochemistry dyeing and Western blottng . [Results] The outcome shows that the group kindled once a day and the group injured with large dose agrypnal have obvious impairment of cognition . In both groups the pCREB decreased and c-fos proteins increased in the hippocamp region of brain .Comparing the two different cognitive impairment causing factors ,the repeted seizure groups showed more obvious retrogression. After the lavage of Caoguozhimutang ,the rats' cognitive function ameliorated greatly ,but did not have distinct difference with the comparison to the control group which was injured with Nimodipine . The pCREB increased and c-fos proteins decreased in the hippocamp region of brain ,which proved the Caoguozhimutang's therapeutic effects on cognitive impairment.
