Artemisinin And Its Derivatives Reversal Of Oral Squamous Cell Carcinoma Cells In Vitro Experimental Study

1 Objective To take the drug resistant human oral squamous epithelium cancer cell(KBV200) as the target cell,to take the arteannuin,dihydroartemisinin,artesμnate and extracts of abrotani herba (crμde extract of alcohol) as the experimental drug,to explore the mechanism of drug resistance of them to tumor through observing its cytotoxic effects on cell,to explore of arteannuin and its derivatives on multi-drug resistance of tumors cells.2 Method To test the cytotoxic effects of vincristine(VCR) and the arteannuin (dihydroartemisinin,artesμnate and extracts of abrotani herba)to KBV200. and calculate the corresponding IC50 throught MTT method ; to observe the arteannuin(dihydroartemisinin,artesμnate and extracts of abrotani herba) concentration at which its inhibition rate to KBV200 is lower than 30﹪,the cytotoxic effect to KBV200 of the arteannuin(dihydroartemisinin,artesμnate and extracts of abrotani herba) accompanied with VCR,to calculate the sensitization index according to the IC50 of VCR plus the arteannuin (dihydroartemisinin,artesμnate and extracts of abrotani herba)3 Results After the experimental study:ⅠUnder the condition of cell culture in vitro,the MTT method experiment showed that the dihydroartemisinin and the artesunate effectively inhibited the proliferation of KBV200,the IC50 were 11.16μmol/l(3.18μg/ml) and1.466μmol/l(0.563μg/ml) respectivelyⅡUnder the condition of cell culture in vitro, the MTT method experiment showed that the reverse rate of the arteannuin at dose of 40umol/l associated with VCR was 3.0 and the reverse rate of the extracts of abrotani herba at dose of 8.88μg/ml and 17.75μg/ml associated with VCR were 2.98 and 12.74 respectively. Meantime,the reverse rate of arteannμin and extracts of abrotani herba correspondingly rise as its density rises.So the arteannuin and extracts of abrotani herba have the effect of reversing the multi-drug resistance of KBV200 cell4 Conclusion The paper proofed firstly that the arteannuin and extracts of abrotani herba have partly the effect of reversing the multi-drug resistance of KBV200 cell and dihydroartemisinin and artesμnate could inhibit the proliferation of KBV200 cell and the inhibitory effect of artesμnate on the proliferation of tumor cell is stronger than of dihydroartemisinin.Results showed: arteannuin and its derivatives have a certain value in experimentary and clinical practice of multi-drug resistance of tumors cells and antitumor...
