Objective: To observe the morphologic changes and the activity of NF-κB , FasL, TNF-α and Caspase-3 in rats retina after exposure to light,and study the protective effect and mechanism of tablet yi — shi against retinal damage by light.Methods: The experiments were performed on 40(Sprague-Ddawley)SD rats which were separated into four groups —normal group, model group , high-dose group and low-dose group.The treated groups received daily tablet yi —shi for 4 weeks.The animals in all kinds of groups, except the normal group, were exposed to green fluorescent light (510~560nm) with an illumination level of 1900+106.9 Lux for 24 hours.After the light exposure the treated groups were given with tablet yi —shi for consecutive one week.Then the pathological changes of retina were examined by light microscopy; and NF-κB , FasL, TNF-α and Caspase-3 were tested.Results: (1) After the light exposure , the outer nucleus layer of retina in the model group became thinner and the regular arrangement of outer and inner segments were disordered, dividing line wasn't distinct. In the high-dose group, the outer nucleus layer of retina got thicker than that of the model group .(2) the level of FasL , TNF-α and Caspase-3 into model group retina increased ,NF-κB decreased, Compared with the normal group ,showed significant different (P<0.01 ~0.05) . In high-dose group , the level of FasL, Caspase-3 and TNF-a obviously decreased , NF-κB increased. Compared with the model group ,showed significant different (P<0.01~0.05) .Conclusion: Tablet Yi-Shi could release the photic injury of retina, accelerate the renovation of retina after light exposure and ameliorate the function of retina.The mechanisms might relate to improve NF-kB level and reduce FasL , TNF-a and Caspase-3 levels so as to inhibit the apoptosis.
