Objective To observe the effect of different kinds of high fat feeding with normal total calorie intake on body fat distribution and insulin resistance and the relationship between them in rats. Methods : Normal male SD rats (6 weeks old ) were divided into three groups : NC (taking normal chow, 18% of calories as fat , n=10);HV (taking isocaloric high-fat diet , 59% of calories as fat which was plant origin , n=10); HF (taking isocaloric high-fat diet , 59% of calories as fat which was animal origin n=10) Each rat was given 310 KJ/d .after 8 weeks ,body weight ,fasting blood glucose ,fasting insulin ,free fat acid ,tissue triglyceride content, epididymal fat pad in rats was observed . Result :There was no effect of high-fat diet on body weight among three groups(HV 308.7±11.65g,n=10;HF 309.7±10.32g,n=10,vs NC 306.7±10.65g,n=10) reflecting the isocaloric administration of the diets, but developed severe visceral obesity.( epididymal fat pad : HV 7.19±0.61g;HF 7.39±0.48g vs NC 4.27±0.93g) Rats taking high-fat diet displayed a small but significant increase in plasma glucose concentration (HV 6.3±0.18 mmol/L; HF 6.4±0.19 mmol/L,vs NC 5.4±0.23 mmol/L)which was associated with slightly elevated plasma insulin levels(HV 16.16±0.52 IU/L,HF 16.16±0.58 IU/L,vs NC 6.9±0.21 IU/L).HV and HF group had significantly higher FFA (HV 780±59.3μmol/L,HF 817±62.65μmol/L,vs NC 512±33.56μmol/L);TG(HV 2.89±0.48 mmol/L,HF 2.96±0.40 mmol/L vs NC 1.1±0.31 mmol/L),compared with CN.HV HF also accumulated triglyceride within the quadricepsmuscle of thigh(HV 7.7 ±0.73μmol/g,HF 7.9±0.70μmol/g,vs NC 4.4±0.57μmol/g) and liver(HV 72.8±0.62μmol/g,HF 74.6±6.13μmol/g vs NC 23.8±4.07μmol/g)relative to CN. Conclusion: As expected, high-fat feeding with normal total colorie intake result in insulin resistance and developed severe visceral obesity in rats. There were no effect of isocaloric high-fat diet on body weight . It showed a positive relationship between insulin resistance and visceral obesity, triglyceride accumulation in tissue, blood triglyceride level .There were no difference between HV and HF group.
