Taraxacum Mogonon(TM), used as vegetable or a Chinese traditional medicine, is a kind of composite and perennial herbage plant. Its flavor is sweet or bitter, and coldness is its characteristic. It has effects of antifebrile and detoxifcation, detumescence and lump dissipation, stomachic, antipyrotic, liver protection and cholagogue, and so on. Many studies found that flavone, amylose, organic acid and terpenoids from natural plants have anti-tumor effect. According to chemical constituent analysis of TM, it has plenty of active materials, such as flavone, amylose, organic acid and terpenoids. Some reports in foreign countries suggested that TM had certain anti-tumor effect, but the studies of anti-tumor effect have not reported in China. Therefore, we explored preliminary anti-tumor effect for TM that was from natural plant to provid some evidences for the developing of anti-tumor drugs or health foods.Part IContent determination of flavone, amylose and chlorgenic acid in extract from Taraxacum Mogonon (ETM)The aim is to study the contents of flavone, amylose and chlorgenic acid in ETM. We used lutin, glucose as criterions and contents of flavone, amylase in ETM were measured by 722 spectrophotometer. In addition, content of chlorgenic acid in ETM was detected by High Performance Liquid Chromatography (HPLC). We also performed quality control, such as the quin precision, recurrence, stability and reclaim rate. The results showed the stability experiments of total flavone, amylose and chlorgenic acid were steady in 1h, 1h and 24h, respectively. All the methods have good precision, recurrence and their reclaim rates were from 95.96% to104.9%. The average content of flavone, amylase, chlorgenic acid in ETM was 5.50%, 5.50% and 1.38%, respectively. The results suggest that ETM has anti-tumor materials such as flavone, amylase and chlorgenic acid whose contents in ETM are reliable. Part II Original studies on ETM's anti-tumor effectThe aim is to explore inhibitory effects of different concentration ETM groups on growth of Hep's hepatoma transplanting tumors in vivo and proliferation of SMMC-7721 human hepatoma cells in vitro. The results showed: In Hep's bearing mice, thymus index was improved in groups of 0.6, 1.2g/kg ETM, tumor weight was decreased in dose of 3.6g/kg ETM and the inhibitory rate was up to 37.07%. The proliferous inhibitory effect of ETM on SMMC-7721 cells was augmented with the increased concentrations and prolonged time (Fig2). The results suggest that ETM has certain anti-tumor effect. Part IIIPrimary studies on mechanisms of ETM's anti-tumor effectThe purpose of this study is to investigate effects of ETM on immune modulation, anti-mutagenesis and their relations with its anti-tumor effect.
