The liver has the function of hematopoiesis. The human fetal liver is a source of hematopoietic a nd immune s tem c ells. H uman f etal l iver has b een u sed t o cure a l ot o f diseases, which hints discovering new protein and new genes correlative with hematopoiesis from the fetal liver are possible, so studying the information of fetal liver's protein may help to understand the mechanisms of liver hematopoiesis. We s tudy t he 2 0 weeks' f etal l ivers o n l evel o f tissues a nd c ells. F irst, w e u sed H E staining to detect the developmental degree of 20 weeks' fetal livers, the result shows the 20 weeks' fetal livers are on the state of developing. Hepatic lobule and hepatic cellular plate have come into being; hepatocytes arrange in single line or double lines and the appearance of hepatocytes are alike; hematopoietic cells are abundant in hepatic sinusoids, staying in the developing stage, a number of red blood cells are found with nucleus. Second, we purified hepatocytes of 20 weeks' fetal livers and acquired the protein expression profile of hepatocytes by two-dimensional gel electrophoresis. It is cells that execute the function of organ, and with cells' physiology state changing, the protein in cells transforms. The hepatocyte is the only parenchymal cell of liver, performing most functions of the liver, which not only participates in metabolism but also forms the microenvironment associated with t he d evelopment o f h ematopoiesis. S o w e purify h epatocytes t o study the information of protein in fetal livers. In our research, we established the technical platform of hepatocytes purification through sedimentation on ice. The purity was analyzed by flow cytometer, indicating that hepatocytes purity was 95.3%. We acquired the protein expression profile of hepatocytes by two-dimensional gel electrophoresis, by refining the lysis buffer and lysis manner, gel preparation, electrophoresis parameters and dying process, we acquired the fine two-dimensional gel maps, and the reproducibility reached to 83%. Our study complemented the data of fetal livers proteomics, laid a foundation of seeking unknown protein and relatively protein of hematopoiesis, settled a cornerstone of researching the liver hematopoiesis mechanism.
