Hypoxia-inducible Factor-1¦Á Expression In Bladder Cancer And Its Significance

Objective:To study the expression and significance of hypoxia-inducible factor-1α(HIF-1α) in bladder cancer .Methods:60 specimens of our hospital normal bladder cancers and 10 specimens of our hospital normal bladders were selected,men 49 case,women 21 case,age:22-85 years old,average 60.6 years old.tumour grade:grade Ⅰ19 case ,grade Ⅱ 30 case,grade Ⅲ11 case.clinical stage:superficial bladder cancer(Tis-T1) 21 case,invasive bladder cancer(T2-T4) 39 case.The expression of HIF-1αmRNA and protein were studied by using tissue chip technique and S-P immunohistochemical method and in situ hybridization.Results:In 60 specimens of bladder cancer patients,the positive rates of HIF-1α protein were 83.3%(50/60).In 19 specimens of grade Ⅰ bladder cancer patients,the positive rates of HIF-1α protein were 63.2%(12/19),all of which were low expression .In 30 specimens of grade Ⅱ bladder cancer patients,the positive rates of HIF-1α protein were 90%(27/30),of whih 4 cases were low expression,23 cases were high expression.In 11 specimens of grade Ⅲ bladder cancer patients,the positive rates of HIF-1α protein were 100%,of which 4 cases were low expression,7 cases were high expression.There was no significanct diference between HIF-1αprotein expression rates of gradeⅠ andⅡ,and of gradeⅡ and grade Ⅲ.however,the marked difference was seen between grade Ⅰand grade Ⅲ(P<0.05).In 21 specimens superficial bladder cancer patients,the positive rates of HIF-1α protein were 52.4%(11/21),all of which were low expression. In 39 specimens invisive bladder cancer patients,the positive rates of HIF-1α protein were 100%, of which 9 cases were low expression, 30 cases were high expression,the marked diference was seen between them(P<0.05).In 60 specimens of bladder cancer patients,the positive rates of HIF-1αmRNA were 91.7%(55/60).In 19 specimens of grade Ⅰ bladder cancer patients,the positive rates of HIF-1αmRNA were 78.9%(15/19),all of which were low expression .In 30 specimens of grade Ⅱ bladder cancer patients,the positive rates of HIF-1αmRNA were 96.7%(29/30),of whih 23 cases were low expression,6 cases were high expression.In 11 specimens of grade Ⅲ bladder cancer patients,the positive rates of HIF-1αmRNA were 100%,of which 2 cases were low expression,9 cases were high expression.There was no significanct diference between HIF-1αmRNA expression rates of gradeⅠ andⅡ,and of gradeⅡ and grade Ⅲ.however,the marked difference was seen between grade Ⅰand grade Ⅲ(P<0.05).In 21 specimens superficial bladder cancer patients,the positive rates of HIF-1αmRNA were 76.2%(16/21),all of which were low expression. In 39 specimens invisive bladder cancer patients,the positive rates of HIF-1αmRNA were 100%, of which 24 cases were low expression, 15 cases were high expression,the marked diference was seen between them(P<0.05).In 60 specimens of bladder cancer,there was significant difference between the expression of HIF-1αmRNA and which of HIF-1α protein(P<0.001).There was no positive expression of HIF-1αmRNA and protein in 10 specimens of normal bladders.Conclusion:In bladder cancer,the expressions of HIF-1α protein and HIF-1αmRNA were related to tumor grade and clinical stage.the higher of the bladder tumor grade,the higher degree of expression of HIF-1αprotein and HIF-1αmRNA.the degree of expression of HIF-1αprotein and HIF-1αmRNA in invasive bladder cancer was higher than which of HIF-1αmRNA and protein in superficial bladder cancer.In bladder cancer,the expression of HIF-1αprotein was positive correlated with which of HIF-1αmRNA.The application of tissue chip is viable to test the clinical specimens efficiently.The method has fleetness,convenience,economy and accurate characteristics.
