Part Ⅰ Experimental Study of Rapamycin for RestenosisObjective To investigate the effects of rapamycin on restenosis after interveneional theary, and analysis the possible mechanism of restenosis prevention of rapamycin. Meanwhile, compare the effects of rapamycin and it with clopidogrel and statins—simvastatin on restenosis. Methods Male New Zealand white rabbits were given cholesterol diet (1% cholesterol 5% lard) for 6 weeks. Balloon injury was performed in the right iliac arteries of the rabbit to make models of restenosis. These rabbits were divided into rapamycin group, followed by injected with rapamycin, and associated group, followed by fed with clopidogel and simvastatin. Two doses schedules in the groups were studied. The proliferation of vascular cells was evaluated by means of immunohistochemistry of the proliferation of cellular nuclear antigen (PCNA). Meanwhile, immunohistochemistry method was used to detect the expression of nuclear factor (NF-κB) and intercellular adhesion molecule-1(ICAM-1). Pathological sections of iliac arteries were observed to estimated the changes of vascular. Histomorphlogy by computer analysis of photomicrogrum. The expression of PCNA, NF-κB and ICAM-1 were analyzed by image analysis. Results Rapamycin treatment significantly inhibited neointimal proliferation. (1) Rapamucin group compared with the injured group:① The lumen area(LA) , the intimal thickness(IT), the intimal area(IA), the ratio of IT/MT and IA/MA of the rapamycin group were significantly reduced (P<0.05), with dose-dependent decrease(P<0.05); ②The expression of PCNA, NF-kB and ICAM-1 were significantly reduceds(P<0.05), with dose-dependent decrease(P<0.05). (2) Rapamycin group compared with associated group:① The LA, IT, IA, IT/MT and IA/MA of the associated group were significantly reduced (P<0.05), with dose-dependent decrease(P<0.05);②The expression of PCNA, NF-κB and ICAM-1 were significantly reduceds (P<0.05), with dose-dependent decrease(P<0.05).Conclusion (1) Given cholesterol diet 6 weeks, make the models of hypercholesterolemia; (2) The experimental restenosis-like model can be established using ovewrstretch balloon to injury the iliac arteries of the cholesterol diet rabbits; (3) Rapamycin could prevent restenosis, to some extent, a conspicuous correlation could be identified between injection dose and the proliferation of neointima ; (4) Treated with rapamycin , clopidogel and simvastatin is better than only rapamycin.
