Objective: To study the mechanism of pharmacological effects on the neuroendocrine network of female climacteric rat of GengNianGuiShen Pill.Methods: Bilateral ovaries of some 3-4 month-old female SD rats were resected to make climacteric, castrated models which were randomly divided' into groups with treatments of high-dose and middle-dose GengNianGuiShen Pill, Fufuchun capsule and placebo respectively. Other 3-4 month-old rats were pseudo-operated to make normal contrast group. For these groups, the effects of different treatments on hypothalamic B-EP,NE,DA,5-HT, 5-HIAA, 5-HT/NE, 5-HIAA/5-HT, and DA/NE, serum E2,FSH,LH,and uterus index were observed For the measurement , Serum E2, FSH and LH were measured by radioimmunoassay. Hypothalamic 8-EP was measured by balance-saturation-assay. NE, DA, 5-HT and 5-HIAA were measured by fluorescence-spectro-assay. Uterus index was measured by tissue-weight-assay.Results: High-dose GengNianGuiShen Pill can increase hypothalamic 8-EP level of castrated rats with P<0.01 and obviously decrease hypothalamic DA level with P<0.05 compared with placebo group. High-dose GengNianGuiShen Pill and Fufuchun capsule can obviously decrease hypothalamic 5-HT level of castrated ones with P<0.05 compared with placebo group. High-dose and middle-dose GengNianGuiShen Pill and Fufuchun capsule can obviously decrease hypothalamic 5-HIAA level of castrated ones with P<0.01,but they have no significant effects on serum E2, FSH and LH. High-dose GengNianGuiShen Pill can increase uterus index of castrated ones with P< 0.05.Conclusion: GengNianGuiShen pill can increase the content of hypothalamic 8-EP and correct disorder of hypothalamic neuromodulator. It has no obvious effect on sex hormones of castrated models, but can increase uterus weight with remitting atrophy.
