Objective To study the effect of Feixin mixture (FXM) on experimental cor pulmonale combined with pulmonary hypertension and investigate its mechanism.Methods Adult male Wister rats were given a single dose of monocrotaline (50mg/Kg) to induce the mold of cor pulmonale. 66 cases were randomly divided into normal control group, mold group, FXM group and nifedipine group (n=11) . Then the rats were treated with gastric infusion of normal saline, FXM or nifedipine in 14th days respectively. The parameters of pulmonary hemodynamics were monitored by Gould3400 polygraph system through the cannulation of polyvinyl tube. Right ventricle and left ventricle added with interventricular septum weight ratio, the lung wet weight and body weight ratio were measured. The level of plasma endothelin-1 was determined by the method of nonequilibrium. The changes of endothelial nitric oxides synthase(eNOS) in rats lung and the small pulmonary arterial morphologic changes were measured by morphmetric and image analysis.Results (l)FXM and nifedipine significantly inhibited the progression of pulmonary artery pressure, decreased the right heart index, and reduced the seepage out of the lung and the level of plasma endothelin-1 (P<0.05 or P<0.01). The effect of high dose FXM group were better than that of nifedipine group(P<0.05 or P<0.01). (2) FXM and nifedipine could inhibit the structural remodelling of small pulmonary arterial and significantly increased the expression of eNOS in endothelia of lung vessels. The effect of high dose FXM group were better.Conclusion FXM is able to reduce the pulmonary hypertension and improved the right heart and lung function. Its mechanisms may be related to reducing the level of plasma endothelin-1 and significantly increasing the expression of eNOS in endothelia of lung vessels.
