Recently piezoelectric crystal biosensors were widely applied in the areas of medical laboratory diagnosis, environment monitor, industrial manufacture, foods sanitary control, etc. Piezoelectric impedance biosensor is simple, sensitive, rapid, low cost, small size and can be used for on-line detection. In this thesis, it was used to determine M.tuberculosis. BACTEC MGIT 960 system was used to select the culture medium that used for detection of H37Rv by piezoelectric impedance biosensor.1. H37Ra was detected by piezoelectric impedance biosensor. The effect of the background conductivity of culture medium and cell constant were discussed. The relationship between the concentration of M.tuberculosis and frequency detection time was determined. Typical frequency response curve for M.tuberculosis was measured. The effects of other Microbes in the stimulation samples were also investigated.2. The culture media that suitable for determining H37Ra could not be used for H37Rv and NIM.TB. An ideal culture medium was found by using BACTEC MGIT 960 system, general tubes and a LGC number resistance bridge. Furthermore, the effects dextrose, fat acids and their derivations were investigated in experiment andtheory.3.Two kinds of stainless steels were used as electrode material. Several passivated methods were used for passivating electrode and the best passive method was selected. Using selected method, a good response curve for H37Rv was obtained by a piezoelectric impedance biosensor.
