Nourishing Formula And Induction Of The El9611 Murine Leukemia Cell Apoptosis And Mechanism Experiments

Objectives: To investigate the effect in acute erythroblastic leukemia mouse EL9611 by Yiqiyangyin formula. To investigate whether the Yiqiyangyin formula induces apoptosis or not. To explore the relation between the effect and apoptosis. To investigate the expression of Bcl-2 and Caspase-3 in leukemia cells. To explore the relation between Bcl-2 and Caspase-3 and leukemia cell apoptosis.Methods:The effect of Yiqiyangyin formula on acute erythroblastic leukemia of EL9611 mouse was observed by white blood cell calculation, reticulocyte calculation and leukemia cell calculation of bone marrow , etc. The bone marrow cell apoptosis was observed by using optical microscope and flow cytometer. Exepression of Bcl-2 and Caspase-3 was observed by immunochemical assay.Results:After the treatment of Yiqiyangyin formular, white blood cell calculation and leukemia cell calculationg of bone marrow of EL9611 mouse was decreased and reticulocyte calculation was increased, etc. The typical feature of apoptosis of bone marrow cell induced by Yiqiyangyin formula was observed by using optical microscope and flow cytometer. Immunochemical results showed that the expression of Caspase-3 was upregulated and the expression of Bcl-2 was downregulated by Yiqiyangyin formula.Conclusion:Yiqiyangyin formula shows good effect in acute leukemia mouse EL9611 . Yiqiyangyin formula induces apoptosis of leukemia cell. The increase of apoptosis of leukemia cell play an important role in the effect. The upregulation of Caspase-3 and the downregulation of Bcl-2 play a fundamental role in apoptosis induced by Yiqiyangyin formula.
