Chinese Medicine Decoction Xiaoji Vitro Study, Ovarian Cancer Chemotherapy And Hyperthermic Chemotherapy Sensitizing Effect

Background & Objective: The drug-resistant of human malignant ovarian neoplasm have significant effect on chemotherapy of human ovarian carcinoma. The objective of this study(1)To investigate the impact of Xiaoji Decoction, Chinese medicinal herb, on proliferation of drug-resistant human epithelial ovarian carcinoma cell line CAOV3/cDDP and the synergetic effect of Xiaoji Decoction and cisplatin, and explore its mechanism. (2)To determine the effect of inhibition of cell growth of hyperthermia in combination with cisplatin on drug-resistant human ovarian cancer cell line CAOV3/cDDP, and the relationship between temperature and medical effect.(3)To evaluate the effect of a concomitant treatment with Xiaoji Decoction, hyperthermia and cisplatin in drug-resistant human ovarian cancer cell line CAOV3/cDDP.Methods: CAOV3/cDDP cell line were treated by Xiaoji Decoction (0.5mg/ml, 5mg/ml, 50mg/ml),hyperthermia (42℃) or cisplatin (6ug/ml, 12ug/ml, 24ug/ml) alone , or by the combination of them . The growth inhibiting rates to human ovarian cancer cell line CAOV3/cDDP were determined by methyl thiazolyl tetrazolium (MTT) method. Signs of induction differentiation were observed by colony forming units assay,respectively.Light microscopy and phase-contrast microscopy were applied to examine the changes in morphology and structure of CAOV3/cDDP cell line, in order to evaluate cell apoptosis.Results: It was found that Xiaoji Decoction combining with cisplatin obviously increased the inhibitory rates and apoptosis rate of the human ovarian cancer cell line CAOV3/cDDP than those of the corresponding individual cisplatin groups (P<0.01). Within a certain concentration range, the synergetic effect of Xiaoji Decoction and cisplatin were significantly different in dose-dependent manner.Xiaoji Decoction in high have synergetic effect with cisplatin. Compared with various concentrations of cisplatin, hyperthermia at 42℃ for 60 min with cisplatin markedly inhibited the cell growth (P<0.01) . When all treatments were given concurrently, the response was greater thari calculated response of all three individual treatments, indicating a synergistic interaction (P<0.01) .Morphological observation indicated that Xiaoji Decoction inducing CAOV3/cDDP death characterized by apoptotic phenotype.Conclusion: Xiaoji Decoction can enhance the action of cisplatin. Its mechanism might relate to induction of apoptosis. Hyperthermia at 42℃ for 60 min combined with cisplatin can suppress growth of human ovarian carcinoma cells. The combination of hyperthermia, cisplatin and Xiaoji Decoction is not only a good combination for potential clinical application, but also an effect way of dealing with cisplatin resistance.
