Shenshu Ii On Adriamycin-induced Nephrotic Syndrome Rat Model Of Experimental Research

Objective:To study theraputic effects and mechanism of shenshu II on Adrianmycin induced nephrotic syndrome(NS) in rats.Methods:Sixty male Wistar rats were selected and randomly divided into two groups.Twelve rats were used as normal group,and the remaining rats were given intravenouse injection of Adrianmycin (6.0mg/kg ) to establish model of NS.The model of NS rats were divided into four groups: the model group(n=12), shenshu II group (n=12) treated with shenshu II alone , shenshu II and prednisone(n=12) treated with shenshu II combined with prednisone, prednisone and persantine group(n=12)treated with prednisone combined with persantine for 21 days respectively. Normal group and the model group were given 0.9% sodium chloride. Three weeks after treatment ,the general condition of rats,24h urine protein quantity, serum albumin,totalalbumin, plasma creatinine, urea nitrogen ,cholesterol , triglyceride, D-Dimer ,GMP-140 and pathological changes of renal tissues were observed.Result:24 hour urine protein quantity increased significantly 7 days after the injection of Adriamycin .After 21 days treatment,groups treated showed significant decreasing in 24 hour urine protein quantity, cholesterol triglyceride, D-dimer, GMP-140, and increasing serum albumin ,total albumin(p<0.01,versus the model group).Shenshu II combined with prednisone got better theraputic effects(p<0.05, p<0.01,compared with the other treatments groups). D-Dimer,GMP-140 in the group treated with Shenshu II decreased significantly (p<0.01,compared with the other treatment groups). Pathological studies showed obvious fusion of footprotrusion without marked thickening of glomercular basement membrance in the model group. The fusion of partial footprotrusion was observed in the groups treated.Conclusion:The results showed:(1)the nephrotic syndrome models were induced by intravenouse injection of Adrianmycin, (2) all treated groups could get the theraputic effects,(3) the theraputic effets in the Shenshu II combined with prednisone were better than the others, (4) D-Dimer,GMP-140 in the group treated with Shenshu II decreased significantly compared with the other treated groups,which suggested the treatment with Shenshu II could get better theraputic effets on nephrotic syndrome by improving the coagulation conditions.
