| The nature phenomena and the human beings life phenomena was united in the Huang Di Nei Jing Canon of Medicine,and formed the theory that human beings and nature are an integer．The theory of Time-Viscera & Yin-Yang is an important proper part of theoretic system of traditional Chinese medicine(TCM)．This subject is divided into theoretic exploration,experimental research and clinical research．Theoretic exploration generalize the theory of Time-Viscera & Yin-Yang in the Huang Di Nei Jing Canon of Medicine first．This theory concentrative express the holistic concept of Traditional Chinese Medicine．It has two main components:1 A human body is regarded as an integral,the theory of the five elements explain that the Five-Viscera Controling System how to integrate human beings' every tissues into a whole system．The mechanism of "Qi" is functional basic of the Five-Viscera Controling System．2 Human being are considered in conjunction with the climatic,geographic,social and other environmental factors,rather than considering the human body as an isolated incident．Second,studys liver and kidney controling system functional characteristic．Dispersing and discharging functions of the liver generalize the function characteristic of the whole liver controling system．The whole kidney controling system's functional characteristic is storing and sealing．There is a very close relationship between liver and kidney．They have same origin and nourish each other in substance while they inhibit each other in function．Kidney keeping in well function of storing and sealing is precondition of modest dispersing and discharging function of liver．When Liver's discharging ＆ Kidney's storing in balance human body will be in steady state,otherwise will be in disorder．Morden medicine research proved that neuro-endocrine-immune network in disturbance while liver and kidney function disorder．Tardive Dyskinesia(TD) is one of the severest side effects of long-term treatment with neuroleptic.There is not TD name in TCM．According to clinical situation, TD can be come under "ChanZheng" concept of TCM. Therer is a close relationship between cause of or therapy for"ChanZheng" and liver and kidney controling system.Based on it,we make out "Chai-Hu-Tao-Ren Compound" to adjust liver's discharging and kidney's storing.The experimental research has two parts:In the first trial,focus on observing the method of high dose haloperidol(2mg/kg/day)abdominal cavity injection induced spontaneous orofacial movements in rats,assessed rats vacuous chewing movements(VCM) in 5 min．Examine SOD (superoxide dismutase)activity in serum at the end of the trial．The result showed that there was significant difference in VCM scores between three neuroleptic-treated groups and controling group on the 40th day(P＜0.05 )．No significant difference was found among three neuroleptic-treated groups(P＞0.05)．The resultrepeated the method of high dose haloperidol-induced VCM as an animal model of TD successful．The TCM group treated with Chai-Hu-Tao-Ren Compound significantly decreased neuroleptic-induced VCM while no significant difference between Clonazepam group and model group．The TCM group had significantly higher SOD activity in serum than the model group and Clonazepam group both．The model group had significantly lower level of SOD than the control group．To study further about oxidative stress as the pathogenesis of TD,chosed vitamine E instead of Clonazepam in the second trial and clinical research．In the second trial,rats were treated with high dose haloperidol(2mg/kg/day) abdominal cavity injection,examine SOD activity,MDA(maliec dialdehyde) level in serum at the end of the trial．TCM group and vitamine E group both had lower VCM scores than pretherapy．Compared with the model group,TCM group's post-therapy VCM scores,reduce scores,rate of reduce scores had significance(P＞0.05)whilie vitamine E group didn't．Model group's serum SOD activity significantly lower and MDA... |
