Syndrome is the center of the clinic dialectic differentiation in symptoms and signs in Traditional Chinese Medicine (TCM). It is based on TCM, especially on Zangxiang theory and directs the clinic practice on prescription. Therefore, the study of TCM syndrome, which is related closely to the Zangxiang theory and the traditional Chinese medicine made of two or more ingredients, is the key problem on the development of TCM theory. According to the clinic investigation, the incidence of a liver disease is 39.56% of all the disease on the five internal organs. Of them, the stagnation of liver-energy and some interrelated syndrome rank the first place, 41. 9% of all the liver disease. Stagnation of liver-energy is the result of the basic pathologic change of liver disease, and is the center of liver syndrome. In the liver syndrome, stagnation of liver-energy and spleen weakness have the largest number with 21. 25% So the study on the formation of stagnation of liver-energy has very important theoretical and realistic significance on the theory of TCM syndrome. To solve the problems, we should develop animal models that are suitable for the characters of TCM syndrome.According to the TCM basic theory and modern mature results on traditional Chinese and western medicine, the paper explained the internal meaning of "liver bears the disperse effect". On the basis of pathogenesis, clinic performance and therapeutical effects, the observation of the wastage of the 1% saccharose, combined with the present realization of the related disease, such as depression, and the effect of Xiaoyaosan, the paper established a big rat model, which was fit for TCM theory on chronic stress stagnation of liver-energy. And through the test of blood plasma ACTH, the BDNF expression on immunity and the original observation of the big rat's pallium, the paper had a research on the formation mechanism. This study not only enriched and created the modern realization on the TCM liver phenomena, and contributes to the TCM clinic prevention and cure on stress, but also provided objective evidence for the further study on the effect of Xiaoyaosan (xys) against depression, even further spread the clinic practice. All of them had higher theory value and greater realistic significance.This paper had two parts: theoretical research and experimental research.The theoretical part began with the function of the "liver bears disperse effect". It put emphasis on the fact that the body's adjusting, blood's circulation, fluid's transport and the spleen or the stomach's cleanliness or turbidness were up to the liver's disperse effects. After that, we discussed the diverse reflections of stagnation of liver-energy due to the stimulation of feelings, and paid attention to the characters stagnation of liver-energy and had a discussion on mechanical analysisand remedial principals. Besides that, we also had a primary discussion on the psychological stress, stagnation of liver-energy and depression, and believed that it was fit to make an animal model by the means of chronic psychological stress. The reasons were: (1) abnormality in feeling was the ma in reason. Because TCM realized psychological stress by feeling disease, it was suitable to the reality by using a chronic psychological stress way to make a model. (2) Stagnation mainly showed psychosis depression, sentimental and susceptible symptom. Chronic psychological stress made the animals' movements land the amount of tasty food less. Such anastomoses to the clinic main symptom; on the other hand, when in chronic stress, the weight of animals increased slowly showed that it was stagnation of liver-energy and weak in spleen. (3) TCM thought that feeling and liver had a close relationship. The liver played a decisive role in the process of psychological stress.
