Objective: The injury and repair of cartilage is a long-term existed problem. This experiment was conducted to observe the effect of cartilage-derived growth factor (CDGF) on the cultured rabbit chondrocytes and its synergistic action with insulin-like growth factor-I (IGF-I). Through it, we studied systematically the relation between CDGF, IGF-I and proliferation and metabolize of chondrocytes, demonstrated the network regulation effect of growth factor on the proliferation and metabolize of chondrocytes, provided a new idea and reliable experimental proof for the treament of cartilaginous disease on the clinic. Materials and methods: CDGF was extracted from chicken xiphoid in our laboratory. Chondrocytes were isolated from 3 weeks old New Zealand male rabbits, cultured in Ham's F-12 medium containing 10% fetal calf serum. The chondrocyts of second generation was treated with CDGF and\or IGF-I of different concentration, through the chloramine T method and MTT method, we compared the content of hydroxyproline in the Ham's F-12 medium, observed proliferation and energy synthesis of chondrocytes. Results: CDGF and IGF-I can both stimulate the proliferation and synthesis of hydroxyproline of chondrocytes dose-dependently, the effect of them on proliferation of chondrocytes had no statistic difference, but the activity of IGF-I on the collagen synthesis of chondrocytes was higher than that of CDGF. They was both active at nanogram level. There was obvious synergic effect between CDGF and IGF-I. Conclusion: CDGF can stimulate the proliferation and collagen synthesis of chondrocytes and has synerg- istic effect with IGF-I.
