AbstractObjective To study the action of anti-atherosclerosis (AS) of the extract of scallop skirt (ESS), and to observe the effect on vascular smooth muscle cells (VSMCs) proliferation of ESS and its polysaccharide-glycosaminoglycan (GAG). Methods ①In order to observe the effect on serum and tissue lipids level and atherogenic plaque of ESS, we established experimental atherosclerotic quails by feeding high lipid forage. ②The inhibitory effects of ESS and GAG on the proliferation of rat VSMCs induced by 20% fetal bovine serum (FBS) or basic fibroblast growth factor (bFGF) were investigated in vitro culture by cell counting, crystal violet staining and MTT assays measuring. At the same time, we observed the effect of ESS and GAG on the expression of proliferating cell nuclear antigen (PCNA) and platelet-derived growth factor (PDGF) of proliferating VSMCs through immunohisto-chemical technique (LSAB method). Results ①Compared with model group, the atherosclerotic plaque of aortic and coronary arterious endothelium in ESS group was significantly decreased (P<0.05,P<0.01). The serum total cholesterol (TC), triglyceride (TG), low density lipoprotein-cholesterol (LDL-C), very low density lipoprotein-cholesterol (VLDL-C), apolipoprotein B (ApoB), and TC and TG in heart and aorta of ESS group were obvious lower than those of model group (P<0.05,P<0.01). ②The number and absorbance of VSMCs in ESS and GAG groups were less than model group (P<0.05,P<0.01). Furthermore, ESS and GAG reversed the enhanced expression of PCNA and PDGF (P<0.01). Conclusion ESS possessed significant anti-atherosclerotic action , which might be related to the adjustment of serum lipids level of ESS and the inhibition of VSMCs proliferation of ESS and GAG.
