Objective To determine the expression of LRP gene in acute leukemia patients and evaluate the relationship between LRP expression ,multidrug resistance and prognosis.Methods Expression of LRP,nm23-H1,MRP mRNA in bone marrow cells were determined from 47 acute leukaemia ,5 chronic granulocyte leukamia patients and 7 normal cases by semi-quantitative reverse transcription polymerase chain reaction(RT-PCR).Results Expression of LRP gene was negative in 7 normal cases, positive in 17 of 25 acute myelocyte leukemia, 1 of 12 acute leukocyte leukemia,8 of 10 refractory or relapsed cases.An analysis demonstrated LRPmRNA level was significantly higher in newly diagnosed acute myelocyte leukemia and refractory or relapsed cases than in normal cases(P<0.05,p<0.05),increased LRPmRNA level has correlation with lower sensitivity to initial chemotherapy and was associated with reduced overall survival rate (P〈0.05).Complete remission(CR)rate was lower in LRP positive patients than that in negative cases(27% vs 80%).The level of LRP was correlation with that MRPmRNA(r=0.44,p=0.009),There was no correlation between LRP and nm23-H1(r=-0.108,p=0.54).Conclusion The expression of LRPmRNA can predict the treatment outcome and prognosis for acute myelocyte leukemia,it was more worse in LRP and MRP linked expression cases,So LRP was an important resistant factor ,determination of LRP and MRP can evaluate the prognosis and help us choose chemotherapy program.
