Objective To explore the relationship of zinc cell apoptosis and immunology indicators in psoriatic epidermis. Methods Apoptosis of keratinocytes in psoriatic lesions and normal skin were measured by terminal deoxynucleotidyl transferase-mediation dUTP-biotin nick end labelling (TUNEL) and flow cytometer respectively; The zinc contents in psoriatic lesion and normal skin were measured by atomic absorption speetrophotornetry(AAS). irnmuno-histochemical technique was employed to study the in situ expression of infiltrating T lymphocytes(CD4 CD8 .. CD25) in the lesional psoriatic skin. Alkaline phosphatase-anti-alkal ire phosphotase (APAAP)method was used to determine the levels of CD4and CD8T lymphocyte ,and double antibody sandwich enzyme-liked immuno sorbant assay (ELISA)method to determine the level of sIL-2R, in peripheral blood of psoriatic patients before and after treatment with Theragan-M. Results The higher frequency of apoptotic keratinocytes was observed in progressive stage psoriatic lesion, and the frequency was reduced in static stage psoriatic lesion and normal skin. The zinc contents in psoriatic lesion were varied in different stages of psoriatic lesion. There was a negtive correlation between the contents of zinc and the percentage of apoptotic keratinoeytes in psoriatic lesion. There was a significantly increased expression of infiltrating T lymphocytes (CD4-. CD8 CD25) in the lesionai skin than in normal skin. The expression of CD4 CD25 T cells in progressive stage lesional skin was higher while the expression of CD8 T cells was lower than in static stage lesion. There was a significant increase of CD4 T cells and CD4/CD8 and decrease of CD8t T cells and sIL-2Rin the peripheral blood of psoriatic patients after Thergan-M treatment compared with that before treatment. Conclusions There may be close relationship between the development of psoriatic lession and apoptosis of keratinocytes. Zinc and immunology indicator in Psoriatic Epidermis may play an important role in keratinocytes apoptosis. There is evidence that Theragan-M treatment has an obvious effect en T-lymphocyte subset and sl7L-2R in peripheral blood in patients with psoriasis.
