| Four cell strains of anti-rabbit IgG monoclonal antibodies are obtained. One of the four monoclonal antibodies -RG48G8, which is against Fc fragment, is coupled to polystyrene tubes. These coated tubes is compared with the tubes coated by the immunoaffinity purified donkey anti-rabbit IgG antibodies. Although the monoclonal solid-phase antibody is not fit for application because of its low affinity, the polyclonal solid-phase antibody, together with the donkey anti-sheep IgG coated tubes, shows a good result in T3 RIA and Progesterone RIA(P RIA). The sensitivity of the coated tube T3 RIA is 0.15ng/ml, CVs are 3.5%-13.0% intra-assay and 9.1%-11.2% inter-assay respectively, the recovery ranges from 103% to 134% and the correlation coefficient with solid microparticle T3 RIA is 0.786. The sensitivity of the coated tube P RIA is 0.17 ng/ml, CVs are 2. 2%-9.4% intra-assay and the recovery ranges from 88.6% to 119%, the correlation coefficient with solid microparticle P RIA is 0.943. |
