Chronic glomerulonephritis is a common disease. Its pathologic factor is immuno-induced inflammation, followed by secondary disturbance of blood coagulation in glomerulus. It develops slowly and become glomerulus sclerosis at last. According to its clinical manifestations, it belongs to such diseases as "edema""consumptive disease""hematuria"and so on in TCM. Its pathogenesis of TCM is deficiency in origin and excess in superficiality. That is both the spleen and the kidney deficiency in origin, dampness, heat and blood stasis excess in superficiality. For many doctor's clinical experience, it is differentiated as Qi deficiency and blood stasis mainly. It occurs because of Qi deficiency and is difficult to be cured because of blood stasis. Tong-Mai decoction, composing of Radix Astragali seu Hedysari, Radix Notoginseng has been used to treat chronic glomerulonephritis in clinic and got good effect. To research the mechanism of treatment, we have been carried out some animal experiments. NIH mice were used to do acute toxicity test in experiment one. The result showed the maximal dose which the mice can tolerate is 312 times of clinical dose. In experiment two, rat glomerulonephritis due to anti-thy-i antibody model was made first, then Qi deficiency and blood stasis model was founded based on it by swimming-leading fatigue method. Rats were divided into Tong-Mai high dose group, Tong-Mai low dose group, Jin-Shui-Bao group, model control group and normal control group randomly, 12 rats in each group. After four-week- treatment, curative effect of every group were compared. The result of experiment two towed Tong-Mai decoction could reduce urinary albumin, urinary fibrinogen degradation products (FDP) and concentration level of urea nitrogen and creatinine in serum, improve indexes of blood rheology, regulate cellular immune ftinction, and lighten pathologic changes of kidney. Besides, the most important result was that it could prohibit IgG from sediment in mesangiurn and prohibit hyperplssia of the mesangium cell and matrix. The curative effect of Tong-Mai high dose group is the best compared with Tong-Mai low dose group and Jin-Shui-Bao group, which showed the relationship between dose and effect. These results indicate Tong-Mai decoction can improve and stabilize renal function, which delay the development of chronic glomerulonephritis.
