Differences Within The Council On The Therapeutic Effect Of Ectopic Endometrium In Mice And Its Immunological Mechanism

Endometriosis (EMs) is defined as the presence of functional endometrial glands and stroma outside the uterine cavity.The endometriosis is a common disease in reproductive age women, the incidence of the disease is about 10～15% and 50% in infertile women, it comes with menalgia. dyspareunia and barren in clinical.The mechanism of the endometriosis is not clear. The theory of retrograde menstruation can not explain the fact that retrograde menstruation happened to 90% of all the reproductive age women, but only a few of them sufferd endometriosis. Recently research notice that the change of environment of immunity in abdominal cavity is considered an important factor in happening and developing of endometriosis.The endometria heterotopic cell reflow to abdominal cavity with the retrograde menstruation, and "allowed" survivaling and growing. The inflammation factors in blood, peritoneal and the tissue were rising, such as the IL-1, IL-6. IL-8, TGF-β1 and TNF-α. which were regulated by NF-κB. The high expression of the inflammation factors plays an important role in the happening and developing of endometriosis.Macrophage is the most important immunocell in abdominal cavity, it plays an important role in endometriosis with the change of quantity, the ability of phagocytosis and the ability of secretion. The changes of the macrophage play an important role in the happanning and developing of the endometriosis.Neiyixiao is compacted of Ligustrazine. Ferulaic acid and Tetrahydropalmatine. This research investigates the effect of Neiyixiao in curing the endometriosis in mouse modle.Objective:To explore the effect of Neiyixiao on the accretion ectopic endometrium of rats and the impact of inflammatory factor in blood serum, peritoneal fluid and ectopic endometrium. To investigate the mechanism of Neiyixiao.To investigate the effects of medicated serum of Neiyixiao on the amount, the ability of phagocytosis and the ability of secreting cytokines of peritoneal macrophages, to investigate the mechanism that Neiyixiao cure the endometriosis in cell level.Method:(1) Observe the estrous cycle of rats. Endometriosis was established by surgical implant of endometrial tissue which belongs to its body in estrus. Get the blest rats modle group to control group, model group, gestrinone group. Neiyixiao 0.18 g·kg-1 group. Neiyixiao 0.09 g·kg-1 group. Neiyixiao 0.045 g·kg-1 group by chance. Before administrte the drug to the EMS model of rats. we analys the volume of endometrial tissue of every group by SPSS13.0. it exhibit no distinguished difference (P>0.05). Administrte the drug for 28 days, measure the ectopic endometrium by vernier caliper. and observe the morphology change of ectopic endometrium by HE staining, weigh the spleen by electronic balance.(2)Administrte the drug for 28 days, anesthesia the rats by aether. The content of tumor necrosis factor (TNF)-a and Interleukin (IL)-1(3 in blood serum and peritoneal fluid was detected by ELISA test, immunohistochemical method was used to detect Interleukin (IL)-6.Interleukin (IL)-8 and transforming growth factor-(31 in ectopic endometrium, Inhibitory-κBa(IKBa) protein in ectopic endometrium were analyzed by western blot.(3) Administrte the drug for 28 days, anesthesia the rats by aether. Administrte Neiyixiao,35 min later, gets the blood of rats, and then prepares the serum of Neiyixiao as common practice. Rat peritoneal macrophage were separated different groups and cultivated as common, and then treated with LPS (but not contol group), give the serum of Neiyixiao from different groups to the macrophages correspondingly at the same time. The secreation of IL-1β,TNF-αwas determined by ELISA test, and MTT assay was used to examine cells proliferation. Neutral red was used to detect the phagocytosis ability of macrophage. Western blot assay was used to examine the expression of IκBαin macrophages of different groups.Result:(1) Neiyixiao 0.045 g·Kg-1,0.09 g·Kg-1.0.18 g·Kg-1 group reduced the volume of ectopic endometrium markedly (P<0.05). improve the morphological characters. Neiyixiao 0.18 g·Kg-1 group raised the spleen exponent of rats model markedly (P<0.05).(2) The IL-6. IL-8 and TGF-β1 in ectopic endometrium was also decreased markedly (P＜0.01). Neiyixiao can increase the I↗Ba in ectopic endometrium markedly (P＜0.01).(3) Neiyixiao 0.09 g·Kg-1.0.18 g·Kg-1 group decrease the TNF-a and the IL-1βin blood serum and peritoneal fluid markedly (P<0.05).(4) The serum of Neiyixiao 0.18 g·kg-1,0.09 g·kg-1,0.045 g·kg-1 has no effect on the cells proliferation (P>0.05), but the content of IL-1β,TNF-a secreted by macrophage was decreased by it (P<0.05). It also increase the expression of IκBα(P＜0.05). The serum of Neiyixiao 0.18 g·kg-1 can enhance the phagocytosis ability of macrophage (P<0.05).Conclusion:(1) Neiyixiao can inhabit the accretion of ectopic endometrium tissue of rats, and its mechanism might be associated with improving the environment of body.(2) The serum of Neiyixiao could inhibit the secretion of IL-1β,TNF-αby LPS-induced macrophages. and the mechanism must be associated with increasing the expression of IκBα. The effect was no due to inhibition of the proliferation of peritoneal macrophage.
