Background and Basis:Depressive disorder is an important factor of progression of breast cancer. Depressive disorder can result in obvious immunosuppression. Recently researches showed that Myeloid-derived suppressor cells (MDSCs) are a heterogeneous population of cells that expand during cancer and inflammation, and that have a remarkable ability to suppress T-cell responses. MDSCs are induced expansion significantly could be a major mechanism for immunosuppression of the tumor-bearing host of depressive disorder. The classical method of antidepressant is supporting vital Qi and relieving Qi Stagnancy in Liver. SFJP formula is a empirical formula that assist to anticancer through supporting vital Qi and relieving Qi Stagnancy in Liver. SFJP formula has an adjustive role of the multiplication of MDSCs. Methods:Clinical observation of 40 patients with Hamilton Depression Rating Scale (HAMD) proves that depressive disorder is a very important factor and a common pattern type that boosts the invasion and progression of breast cancer.80 of 112 BALB/c mice were intervened by chronic unpredictable mild stress (CUMS) animal model of depressive disorder. The effect was verified by the tests of ethology. Then the mice were injected with 4T1 subcutaneously and intervened in groups, such as the simple depressive disorder group (A), the group of tumor-bearing mice of depressive disorder with no drug intervention (B), the group of tumor-bearing mice of depressive disorder with Gemcitabine chemotherapy (C), Chinese medicine SFJP formula group (D), the chemotherapy plus Chinese medicine group (E), the simple tumor-bearing mice group (F), and the normal mice group (G). Measured the weights and the major axises of the tumor of mice in twice of every week. After 28 days, measured the weights of the tumors and the spleens of mice in groups, acounted the inhibition rate of tumor, and assayed the MDSCs and subsets, CD3/CD8, CD3/CD4, the level of apoptosis of CD8+T cells in the splenocyte suspension by FCM, respectively.Assayed the IL-6 and the IFNγin the peripheral blood by ELISA.Assayed the NO in the splenocyte suspension by Nitrate reductase.Observed survival in groups.Results:Clinical observation of 40 patients with Hamilton Depression Rating Scale (HAMD) showed that the patients of Depressive disorder before taking breast cancer accounted 72.5%.Confirmed Depression accounted 2.5%. The all patients of Depressive disorder accounted 75%.On the other side, the breast cancer patients who getting Depressive disorder accounted 60%. Confirmed Depression accounted 7.5%. The all patients of Depressive disorder accounted 67.5%%. In vivo, the results of weight of tumor showed the Group B had the greatest weight of tumor.Group B>Group F> Group D> Group C> Group E (28 days and 33 days).Group B> Group D>Group F> Group C> Group E (38 days).There was the highest inhibition rate of tumor and the best survival consisted in Group E.It is showed that the Group E acquired the best effect. Group B had the maximum weight of spleen, while group A had the minimum.The amount of GRldimCD11bbright (MDSC) (%) and CD8+T cells (%)of group B,C,D,E were 25.05%,15.27%,21.06%,11.78%, and 4.86%,6.5%,6.35%,7.47%, respectively.Group B had the maximum MDSCs of spleen. It contained mainly CDllb+LY6G+LY6Clow. The amount of CD3/CD4 (%)of group B,C,D,E were 16.94%,21.78%,19.42%and 26.46%, respectively.Group B had the minimum CD4+cells of spleen.The amount of apoptosis of CD8+T cells showed significant differences between group B and the group E. Annexin-V/PI(%) of group B, C, D, E were 4.63%,3.30%,2.48% and 3.04%, respectively.The highest level of apoptosis was Group B.However, Group E less than Group B,C,F in the amount of apoptosis of CD8+T cells. Furthermore, the group B had the highest level of IL-6. Conclusions:Depressive disorder boosts the invasion and progression of breast cancer and the large expansion of myeloid-derived suppressor cells to a great extent.It is showed that the immunity of the mice of simple depressive disorder (Group A) is suppressed obviously. Depressive disorder with tumor can boost progression of breast cancer. Besides, MDSCs have a remarkable ability to suppress T-cell responses through leading to apoptosis of CD8+T cells. So MDSCs are induced expansion significantly could be a major mechanism for tumor progression. In addition, it is showed that the effect of tumor inhibition in the group of tumor-bearing mice of depressive disorder intervened with chemotherapy plus Chinese medicine (E) is considerably significant.Chinese medicine SFJP formula used simply and combinated Gemcitabine chemotherapy can suppress progression of 4T1 murine mammary cancer cells and prolongate the survival.It is showed that MDSCs are suppressed, the level of NO is reduced and apoptosis of CD8+T cells is slowed down could be the major mechanism for the effect of SFJP formula.
