Lung being relative to the large intestine is one of the most representative theory of zang-fu viscera theory. And the theory is widely used in clinic. The bronchial asthma is a kind of immune allergic disease caused by many factors, which is featured by reversible airway obstruction, airway hyperreactivity and airway inflammatory. According to the world health organization reports, asthma has become one of the global problems which is seriously harm human health. Besides airway inflammatory theory, immunization and allergy doctrine and genetic factors, neural control mechanism is also play an important role in asthma pathogenesis. And the lung neuropeptide mediated neurogenic inflammation is also valued.Based on the literature review and lung being relative to the large intestine theory, the object of this project is to observe the influence of Da Cheng Qi decoction on neuropeptide in lung and intestine of asthma rats through experiments coping asthma mice model. This paper can be divided into three parts:Part one Literature reviewThis paper review the modern research progress of neuropeptide and bronchial asthma, the understanding of bronchial asthma and features of treatment based on syndrome differentiation of TCM.Modern research proves that neuropeptide plays an important part in regulation of breathing and has a closely relationship with symptoms pathophysiological of asthma. And the experiment results show that some Chinese native medicine preparation can adjust the neuropeptide of asthma mice model. At present some neuropeptide receptor antagonist was published, and its role in prevention and treatment of asthma had made the preliminary progress. TCM has made great progress in treatment of asthma by using its holism conception, visceral doctrine and diversified treatments. It thinks the etiology and pathogenesis of asthma more tend to pathological factors of sputum primarily, which is caused by more than exogenous evil. And its main pathogenesis is that the sputum osbtructs air way and makes the lung lose the regulate function of Qi.Part two Theoretical researchThis paper discuss the theory foundation of lung being relative to the large intestine theory from the aspects of the position of lung and intestinal, modern research, physiology, pathology and treatment. And discuss the application and action mechanism of Da Cheng Qi decoction in the treatment of asthma. Lung and large intestine have closely relationship in physiology and pathology. And their clinical treatment can be divided into lung disease to treat bowel, bowel disease to treat lung and the treatment of lung and bowel at the same time. Studies have found that asthmatic patients on attacking stage mostly have stool abnormal symptom. Based on the lung being relative to the large intestine theory, we can make definite effect in the treatment of asthma caused by phlegm-heat accumulation in the lung through lowering the adverse Qi and eliminating sputum. In the discussion of asthma caused by heat accumulation and lung Qi failing to bear downward in Treatise on Febrile and Miscellaneous Diseases, Zhang zhong jing thinks, through lapactic, we can use Da Cheng Qi decoction to treat asthma caused by heat accumulation in the bowel of excess syndrome of YANG MING FU-organ. So modern physicians using Da Cheng Qi decoction to treat asthma is both the inheritance and development of typhoid theory and the clinical application of lung being relative to the large intestine theory.Part three Experimental studyObjective:The prevention and cure function of Da Cheng Qi decoction to asthma mice was studied by observing the physiology and pathology changes of laboratory mice lung tissue. And the changes of TFF3, NKA and VIP neuropeptide were analysed to study the regulation function of Da Cheng Qi decoction to asthma mice. The relationship between the regulation function and content changes of neuropeptide was further studied too.Methods:The bronchus asthma models were established using the complex forms of OVA sensitization and inspiration and the mice were divided into four groups, normal group, model group, normal with medicine given group and model with medicine given group. In the seven days from 14 to 20, normal with medicine given group and model with medicine given group were given Da Cheng Qi decoction through intragastric administration and normal group and model group were given water. The physiological and pathological change of mice lung tissue was observed by HE staining and cells were counted and classified using Wright's staining. Meanwhile, the content of TFF3, VIP, NKA neuropeptide in the mice lung tissue were detected.Results and analysis:(1) Compared with normal control group the atomization inhales in the OVA process, the stimulation tempts breathes heavily when may observe the model group mouse to present the fierce cough, to sneeze, symptoms and so on scant of breath, respite, the partial mice may hear and pant noisily obviously the sound.(2)Compared with model group, the model presents the polypnoea, the musculi abdominis for the medicine group mouse part to twitch, positive reactions and so on feces and urine incontinence, but the degree compares the model group to reduce.(3) Compared with normal control group through microscopic, model group's alveolar walls have congestion and a few RBC, the bronchial epithelium become thinner and part of them fell off, endobronchial and its surrounding have obvious inflammatory cells infiltration, a few epithelial cells and RBC fell off in alveolar. These were similar to the pathological changes of lung tissure of airway hyperreactivity patients.(4) Compared with model group, model with medicine given group's alveolar walls without congestion, endobronchial and its surrounding have no obvious inflammatory cells infiltration, bronchial epithelium was complete. It reveals that Da Cheng Qi decoction has good improvement action on the pathologic changes of asthma mice.(5)Compared with normal group, the amount of lymphocytes, eosinophils and neutrophils of BALF were higher in model group and it has significant difference (P<0.01). And the growth of BALF eosinophilic was the most obvious. It reveals that the inflammation with eosinophils increase mainly was occurred in mice airway of model group.(6)Compared with model group, the amount of eosinophils, neutrophils and lymphocytes of BALF in model with medicine given group were significantly reduced and it has significant difference (P<0.05). It points out that Da Cheng Qi decoction can significantly reduce the percentage of eosinophils, neutrophils, lymphocytes and inflammatory cells and restrain the infiltration of airway inflammatory cells. And this reveals that Da Cheng Qi decoction have anti-inflammatory effects.(7) Compared with normal group, in model group, the content of NKA in lung tissues has rising trend while the content of VIP and TFF3 have trend to decrease.(8) Compared with model group, in model with medicine given group, the content of NKA in lung tissues has a different degree of trend to decrease while the content of VIP and TFF3 have rising trend.(9) Compared with normal group, in model group, the content of NKA in bowel tissues has rising trend while the content of VIP and TFF3 have trend to decrease.(10) Compared with model group, in model with medicine given group, the content of NKA in bowel tissues has a different degree of trend to decrease while the content of VIP and TFF3 have rising trend. From the experimental results, we can see that Da Cheng Qi decoction can reduce the release of inflammatory cells and significantly regulate the content of NKA, VIP, TFF3 in the lung and bowel tissues of mice. It reveals that Da Cheng Qi decoction can regulate the inflammation of lung tissue and the content of neuropeptide in lung and bowel tissues. The research explains the mechanism of using Da Cheng Qi decoction to treat asthma in certain degree and provides experimental basis for lung being relative to the large intestine theory.Conclusion:In conclusion, Da Cheng Qi decoction has good improvement action on the pathologic changes of asthma mice lung tissues. It can also alleviate or prevent asthma attack through lowering the adverse Qi and regulating Qi activity of lung and bowel tissues to reduce inflammatory cells infiltrating of asthma and to regulate the content of NKA, VIP, TFF3 in lung and bowel tissues. And these function may be one of the mechanism of lung and bowel effect of using Da Cheng Qi decoction to prevent and cure asthma.
