Experimental Study Of Gynecological ¢ò Treatment Of Female Mice Reproductive Tract Ureaplasma Urealyticum Infection

OBJECTIVE:This study focuses the mice model of pelvic inflammatory disease infected by ureaplasma urealyticum serotype;to obsersve the curative effect of the prescription FukeerhaoFang on the young adult female mice with UU infection; and to provide effective treatment of Chinese medicine for clinical treatment of UU infection of the female reproductive tract'.METHEDS:Oestradiaol-treated female BALB/c mice were inoculated intravaginally by serotype 8 of ureaplasma urealyticum. Ureaplasmas in the genital tracts were cultured 3 weeks after inoculation. UU was successfully inoculated. We divided them randomly into two groups:seperatedly given FukeerhaoFang and Azithromycin. After two weeks, vaginal discharge was got by a nasophar-yngeal swab and inocullated into the liquid medium; Histopathologic changes of vagina, cervix, uterine and oviduct of mice were examined by light microscopy;ureaplasma urealyticum were detected by nested polymerase chain reaction; and to observe the distribution of icam-1 in genital tracts.RESULTS:l.An animal model of genital tract infection of Ureaplasma urealyticum was established successfully.2.The infection can spead to the upper genital tract. Genital tract occurs with acute and chronic inflammatory.changes.The overall status of the mice infected with UU is worse.3.Cultivatio:The total positive rates of UU8 in three experimental groups showed a difference, which were 80%(FukeerhaoFang),100%(Azithromycin).The study indicated that both the two thera-peutics have curative effect, but the Azithromycin is little better than FukeerhaoFang. Nested polymerase chain reaction:The results showed that:After treatment,the number of western medicine group is more than traditional Chinese medicine group in genital mycoplasma negative cases, but the difference was not statistically significant.4.Average optical density of ICAM-1 measured using analysis software IPP6 showed that FukeerhaoFang is better than Azithromycin treatment group in the oviduct of mice.CONCLUSIONS:1. The animal model of genital tract infection of Ureaplasma urealyticum can be established successfully.The infection can lead to the PID. It can provide an important basis for animal model of PID.2. Traditional Chinese medicine preparation FukeerhaoFang can significantly improve the state of the mice infected with UU,and decrease the high icam-1 levels caused by UU infection in oviduct. Genital mycoplasma infection can cause the changes of local and general immune function.The differences of the indexes between the two groups tell us the mechanism of genital mycoplasma infection acting on the immune system.It can improve the changes of local and general immune function and eliminate UU.
