Composite recipe of traditional Chinese herbs is the dominant form of the Chinese clinical medication, and the theory of concerted application is the essence of the treatment based on the syndrome differentiation. Compound traditional Chinese recipe, the important composition of the Traditional Chinese Medicine, is effective recipe as well as the essence of the TCM according to the rules of the treatment based on syndrome differentiation and the system of theory,methods,formulas and medicinal and thousand years of clinical therapeutic experience. The research about the pharmacodynamic material basis and functional mechanism in traditional Chinese compound recipes is important to the modern explanation of the theory of concerted application, also it can provide a basis for guidance of the clinical medication and development of new drugs. When a prescription is planed to design, the condition of disease need to be considered and proceeding of design should be strictly abide by the principle of " sovereign,minister,assistant and guide" on the basis of the theory of treatment and syndrome differentiation, then we can do such aspects of distinct of the primary and secondary, comprehensive consideration and enhancement of the curative effect. In this thesis, We chose "San wei tan xiang san" as the research object, gallic acid (GA) and protocatechuic acid (PA) were selected as research index. The influence of guide herb (sandalwood) was revealed through investigating the pharmacokinetical (pk) changes of "San wei tan xiang san". The main contents are as follows:1. A HPLC method for analysis of GA and PA simultaneous in rats plasma and in kidney,Heart,spleen brain,lung,liver were established. The method validation are in line with the U.S. FDA standard testing of biological samples. The results above indicated that this method was suitable for the investigation of GA and PA in rats plasma and in organ.2. After intragastric administration of GZ-RDK (Guangzao-Rou Doukou) and SWTXS (San wei tan xiang san), the pk parameters of GA and PA in plasma of normal SD rats and Stagnancy of Qi and Blood Stasis (SQBS) model rats are compared the pharmacokinetic parameters. The results indicated that GA and PA were Compartment open model; The sandalwood could accelerate the absorption of gallic acid, slows down the elimination and promote the distribution of the organization; The sandalwood slows down the distribution and metabolism, extend duration of action of drug in the body, and improve the bioavailability. These results shows the synergistic mechanisms of sandalwood in Composite recipe.3. Using proposed liquid method, tissue distribution of GA and PA is investigated to study the influence of synergism between GZ-RDK(Guangzao-Rou Doukou) and SWTXS (San wei tan xiang san). The results shows that after the compatibility of sandalwood, the distribution of GA in Heart is increased; the distribution of PA in lung is increased. This conclusion is to provide some evidence theory that sandalowood belong to Spleen, stomach, heart, lung.4. With the change of concentration-time curves, Using spss correlation analysis, NM23.0,ES25.0,NM40.0,NM45.0,NM47.6,NM55.5 were ascertained to be the bioactive substances of "San wei tan xiang san". An initial qualitative analysis on them was carried out by HPLC-MS, then the fragmentions corresponded with them were supposed to be:m/z 257.1,m/z 232.4,m/z 294.5,m/z 294.5. A new mode of drug to investigate the bioactive substances of Composite recipe compatible herb was established in vivo studies.
