Objective:Studies have shown that through the cell cycle process of block and intervention triptolide can cause cell growth arrest and play its anti-tumor effects. Since lower concentrations of triptolide can inhibit the proliferation, and by morphological and flow cytometry to further confirm its HEC-1B cells proliferation inhibition may interfere with cell cycle progression through induction of apoptosis. This study was to select human endometrial cancer cell line HEC-1B as subjects, and investigate the effect of triptolide on inhibition to the proliferation of the cell lines and whether apoptosis-inducing effect of signaling pathway activation PI3K/PKB. In recent years, for tumor cell apoptosis, PI3K/PKB pathway in other diseases have a more clear understanding, as to the pathway for the entry point to observe the TP under the action of endometrial cancer cell proliferation and apoptosis, the drug's clinical applications and further interpretation.Methods:Cell culture,the HEC-1B cells were cultured in nutrient solution of DMEM including 10% FBS, 100U/ml penicillin and 100U/ml streptomycin, and then placed in 37℃,5% CO2 incubator. Cultured solution was changed every 24-48h, and before exchange of medium cell growth was observed. Use 0.25% trypsin digesting and subculturing. Cells growth were in monolayers, and chose cells in logarithmic growth phase.Functional studies:Detection of cell proliferation by MTT. The MTT colorimetry method was used to detect the inhibitive effect of Triptolide on cell proliferation; Western blot was used to observe phosphorylated AKT expression of the cell used different TP concentrateon (10ng/ml,20ng/ml,40ng/ml,80ng/ml) and blankResults:(1)Result of MTT shows PI3K/PKB pathway inhibitor can inhibits the growth of HEC-1B distinctly. The inhibit effect is related to the drug concentration. (2)Western blot shows the expression of phosphorylated AKT is related to the drug concentration.Conclusion:Triptolide inhibits the proliferation of HEC-1B. The expression of the cell AKT phosphorylated protein is downtrend while the deth of drug is raising.It shows that the inhibition effect is related to the signal transduction pathway and the take the important effect.The overall tendency of the cell vegetation inhibition ratio used of TP is reinforce when the time is going on.Obviouserly, PI3K/PKB is just one of the way to the HEC-1B multiplication.
