Objective:To investigate a kind of SD rat model of hindlimb ischemia for research . Method: According to different operating methods 38 SD rats were divided into three groups, sham operation group (A group of 12), abdominal aortic ligation group (B group of 12), plus bilateral ligation of abdominal aortic and abdominal perineal artery group (C group of 14), respectively. 6mg/100g ketamine administered by intraperitoneal injection of anesthesia, per abdomen operation to ligate different blood vessels severally according to the previous design. Observated operated animal,s hinds movement, skin temperature, purple and pale, limb necrosis, etc, afteroperation. Afteroperation2weeks, 4weeks, anesthetized rats punctured iliac vein for blood gas analysis per abdomen and get done hindlimb skeletal to HE stain. Results: in afteroperation 3 days,two rats of Group C dead and the rest of animal survival, group A were not obviously abnorm appearance. Aferoperation, Group B and Group C immediately presented hind pale, then cyanosis, cold skin temperature, hypofrontality of limb motion. Above-mentioned symptoms has improved gradually after two weeks, Group B completely recuperation afteroperations 4 weeks. Group c were obviously ischemia limbs such as cyanosis, cold, not full of limb aferoperation, and still remained above-mentioned symptoms in four weeks,each group had no hindlimbs necrosis. There are Statistical difference in the limbs of above-mentioned symptoms (P<0.05) two weeks, and hindlimbs of muscle strength grade had no Statistical difference after four weeks. Venous blood gas of the A B C group are 54.00±3.33,48.17±2.14,34.50±2.88(mmHg), two weeks respectively. 53.33±1.63,51.67±1.21,43.00±2.00(mmHg),four weeks respectively. There are Statistical difference between three groups in two weeks, and group C with A, B too in four weeks, no Statistical difference between group A and B four weeks(P=0.100).Group A presented normal striated muscle structure, group B coloretur unevenness,swelling and distension, muscle of transverse striation elimination, skeletal muscle cell nucleolus deeply stained two weeks and no difference with the normal group four weeks. Group C present skeletal muscles decoration unevenness, cells swelling and distension, muscle of transverse striation elimination, skeletal muscle cell nucleolus stained deeply and intensively, intercellular space widening, but no obviously necrocytosis. Conlusion: the method of bilateral ligation of abdominal plus aortic abdominal perineal artery can make SD rat model of hindlimb ischemia stably.
