| Objective:To screen effective chemical groups of relievinging cough, expelling phlegm and anti-inflammatory in seed of Nigella Glandulifera. Methods: Grease and non-grease were separated by SFE-CO2. Volatile oil were extracted by hydrodistillation. Flavonoid glycoside were separated by ethanol, purifed by resin. Analysis on chemical components of grease and volatile oil by GC-MS. Determine of linoleic acid and thymoquinone by GC. Determine of flavonoid glycoside by UV. Analysis on kaempferol by HPLC. Cough induced by ammonia solution, phenol red excluded from trachea of mice and swelling of ear induced by xylene. Control groups divided into negative, positive and water solution group. Therapeutics groups divided into grease, non-grease, volatile oil and flavonoid glycoside group. Results: Grease, non-grease, volatile oil and flavonoid glycoside were separated. Their percentage were 30%,70%,0.3%,1.52% respectively. Linoleic acid percentage was 39.75% in grease. Thymoquinone percentage was 1.58% in volatile oil. Kaempferol was 0.330mg/g in seed of Nigella Glandulifera. To compare with water solution groups :grease, flavonoid glycoside high dose and volatile oil low dose can prolong patent period.To compare with positive groups:non-grease low ,high dose and volatile oil high dose can decrease the times of cough. To compare with water solution groups : flavonoid glycoside high dose can increase phenol red in trachea. To compare with positive groups: grease low dose can prevent swelling of ear induced by xylene. Conclusion: Grease,non-grease and volatile oil can relieve cough. Non-grease and favonoid glycoside can expell phlegm. Grease can prevent inflammatory. Mechanism research of relievingng cough,expelling phlegm and anti-inflammatory can be guided by the results. Meanwhile resource of Nigella Glandulifera in Xinjiang can be widely developed. |
