Objective:To investigate the effects of propofol on cell viability and apoptosis in developing cortical neurons.Method:Primary cultured cortical neurons from Sprague Dawley rat of 0-24h-old were cltured for 6ds.There were 2×105/ml neurons.The study include three tests.First,neurons were cultured in 96-well plates and divided into 6 groups.There were 4 wells in each group.Group C1 received no treatment;in P1,2,3,4,5 propofol was added at the concentration 2,5,10,20 or 50μg/ml respectively. For 12,24,48h,cell viability were determined by the colorimetric MTT assay.Second,There were 4 wells in each group;groupC2 received no treatment;in group Pⅰ,ⅱ,ⅲpropofol was added at the final concentration of 10,20,50μg/ml respectively.The neurons were incubated for 24 hs.The apoptosis rate was detected using TUNEL cell staining.Third, There were 5 wells in each group;groupC3 received no treatment;in group PⅠ,Ⅱ,Ⅲpropofol was added at final concentration of 10,20,50μg/ml respectively,the neurons were incubated for 24 hours. Westen-blot assay detected the expression of cleaved-caspase3.Results:(1) Cultures treated for 12 hours showed nodetectable effect on neuronal viability.But cultures treated for 24 or 48 hs with high concentrition showed a dramatic decrease. (2) Incubated for 24 hours,using TUNEL assay,propofol treatment significantly increased the apoptosis rate of neurons in a concentration dependent manner.(3) Propofol incubation for 24h up regulated Cleaved-caspase3 of neurons in a concentration dependent mannerConclusion:Propofol treatment with long time and high concentration significantly decreased the cell viability and promoted the cell apoptosis.These results offered guiding functions to the children anesthesia.
