Objective:To investigate the effects of ATP-MgCl2 in neonatal rats with hyperoxia induced lung injury.Methods:Select 2-day-old and 5-10 grams of rats120, were randomly divided into 3 groups (each group n= 40),Ⅰ, the air control group;Ⅱ, hyperoxic model group;Ⅲ, ATP-MgCl2 treatment group. Air control group was exposed to room air, other groups were exposed to 90%O2 for 14 days. The treatment group received intraperitoneal injection of ATP-MgCl2(45 mg/kg) everyday, the air and model groups received intraperitoneal injection of equal normal saline. Recorded the mortality on 3th,7th and 14th,and ten rats in each group were executed and lung tissues were taken out and pathological changes of lung were observed under a light microscope. The expression of TNF-α,TGF-β1,Vimentin were detected by immunohistochemistry.Results:With the accrescence of the high-oxygen exposure time, the model group and the ATP-MgCl2 treated groups showed a higher mortality trends. However,the mortality of the ATP-MgCl2 treated group had a slow increase, the model group had a rapid increase after exposed 7 days in high-oxygen and had a significant difference compared with the treated group(P<0.05,P<0.01).There was almost no expression of TNF-a positive cells in air group, but in the model group the expressional intensity level was gradually enhanced with the accrescence of the high-oxygen exposure time. The expressional intensity level of TNF-a in the ATP-MgCl2 treated group was lower than that of the model group in each time segment. It had significant differences among them (P<0.01).There was a little expression or no expression of TGF-β1 positive cells in air group, but in the model group the expressional intensity level was gradually enhanced with the accrescence of the high-oxygen exposure time. The expressional intensity level of TGF-β1 in the ATP-MgCl2 treated group was lower than that of the model group in each time segment. It had significant differences among them (P<0.05,P< 0.01).There was a little expression or no expression of Vimentin positive cells in air group,there is no significant difference between model group and air group exposed to the high-oxygen for three days.the expression in the model group was starting to be enhanced after being exposed to the high-oxygen for seven days,and reached the peak on the fourteenth day. The expressional intensity level of Vimentin in the ATP-MgCl2 treated group was lower than that of the model group on the seventh and fourteenth day. It had significant differences among them (P<0.05, P<0.01).Conclusion:ATP-MgCl2 has protective effects on lung injury induced by hyperoxia in neonatal rats and decrease the occurrence of bronchopulmonary(BPD).
