| Background and aimsGastric cancer is one.of the most common malignancy the digestive system in adults,which is the leading cause of cancer death worldwide.The main reason for death is invasion and metastasis of tumor.Though studies on gastric cancer have lasted for long time,no great progress was made.So,the problem researchers concentrated on is how to diagnose gastric cancer in early stage and monitor its invasion and metastasis.With the development of studies on laminin receptor,more and more people focused on 67LR and galectin-3.Studies have shown that laminin receptor can be divided into integrin laminin receptor and non-integrin laminin receptor two categories,but non-integrin laminin receptor is stronger affinity than the type of integrin receptor,and it gradually has become the hot research.Non-integrin family of nine is mainly sugar-binding proteins,combined with the glycation LN,which 67LR and Galectin-3 are regarding as two most important non-integrin laminin protein receptors.The expression of 67LR in the tumor cell surface is rich and has a stronger adhesion with LN,which plays an important role in invasion and metastasis of tumors.Galectin-3 combined with galactoside mainly,also known as galactoside binding protein or galactose lectin.A variety of tumor cells express galectin-3,which can interact with the micro-environment,and can change normal stromal cell surface conformation in the extracellular matrix.Galectin-3 can be a substrate of MMPs,which its cleavage fragments also promote tumor invasion and metastasis.Some research has shown that Galectin-3 epitopes may be involved in the formation of 67LR,but the view is still controversial.At present we have not seen the explicit report about the change of Galectin-3 and 67LR in the occurrence and development of stomach cancer,and their relationship research.In this study,to investigate the expression of 67LR mRNA and protein, Galectin-3 mRNA and protein in human gastric adenocarcinoma and their relationship with clinical pathological features.To explore their roles in carcinogenesis and progression of gastric carcinoma and to evaluate their clinicopathologic significances and the clinical practice value.Materials and MethodsWe examined the expression levels of 67LR and galectin-3 protein by immunohistochemical staining with the monoclonal antibody 67LR and galectin-3,and correlated this with clinicopathologic parameters in 82 cases of primary gastric adenocarcinoma.The expression of 67LR mRNA and galectin-3 mRNA was detected with in situ hybridization in 82 cases of primary gastric adenocarcinoma.We randomly collected gastric tissues from about 20 patients as the controls which are gastric mucosas from the distant cutting margin.Results1.The 67LR mRNA expression was detected mainly in gastric adenocarcinoma.The positive rate of 67LR mRNA was higher in human gastric adenocarcinoma(58.5%),compared to normal mucosas from the distant cutting margin(25%).The difference was significant(P<0.01).The positive expression rate of 67LR mRNA was significantly related with differentiation classification,depth of tumor invasion,lymph node metastasis and pTNM stage(P<0.05).2.The 67LR protein expression was detected mainly in gastric adenocarcinoma and little in normal cells.The positive rate of 67LR protein was higher in human gastric adenocarcinoma(59.8%),compared to normal mucosas from the distant cutting margin(30%).The difference was significant(P<0.01).The positive expression rate of 67LR was significantly related with differentiation classification,depth of tumor invasion,lymph node metastasis and pTNM stage(P<0.05),but it did not correlate with sex,age and Lauren classification(P>0.05).3.The positive rate of Galectin-3 mRNA(91.1%) in gastric carcinoma were higher than in normal mucosas from the distant cutting margin(67.6%).The difference was significant(P<0.01).The positive expression rate of Galectin-3 mRNA was significantly related with depth of tumor invasion and lymph node metastasis(P<0.01).4.The positive expression rate of Galectin-3(84.1%) in gastric adenocarcinoma were higher than in normal mucosas from the distant cutting margin(35%).There was significant difference between two groups (P<0.01).The expression of Galectin-3 was significantly related with the depth of tumor invasion,lymph node metastasis and pTNM stage(P<0.01),but it did not correlate with sex,age,Lauren classification and differentiation(P>0.05).5.A positive correlation was found between the expression of 67LR mRNA and 67LR protein(r=0.558,P<0.01).A positive correlation was found between the expression of Galectin-3 mRNA and Galectin-3(r=0.478,P<0.01).6.A positive correlation was found between the expression of 67LR mRNA and Galectin-3 mRNA(r=0.318,P<0.01).A positive correlation was found between the expression of 67LR protein and Galectin-3 protein(r=0.309,P<0.01).Conclusion1.In gastric adenocarcinoma,the 67LR and Galectin-3 protein expression are regulated at the transcriptional level.The elevation of the 67LR and Galectin-3 protein expression is correspondingly the result of elevation of transcriptional level.2.67LR and Galectin-3 mRNA and protein are closely related to the invation and metastasis of gastric adenocarcinoma.3.67LR and Galectin-3 mRNA and protein can be considered as important markers in estimating of biological speciality of gastric carcinoma,and may be a promising therapeutic target for gastric cancer. |
