Pyrophosphate is the intermediate in many kinds of metabolic reaction and it plays an important role in life process. In daily life, pyrophosphate is mainly used as water softenert and food additives. Therefore, the analysis of pyrophosphate is very important.The detection of immunoassay, DNA hybridization and SNP(single nucleotide polymorphism) analysis plays a very important role in life sciences. They have been widely used in biochemistry, medical diagnostics. After the radioactive assay and enzymatic assay, a method of chemiluminescence using metal nanoparticles as probes has been revealed. By detecting the metal ions from metal nanoparticles, we quantitative detect the immunoassay, DNA hybridization and SNP analysis and the result is satisfactied with high sensitivity. Because the usual metal nanoparticles as Au, Ag are hard to be dissolved into metal ions, the metallic oxide-Fe3O4 easily dissolved has been prepared and the results of Fe3+, Fe2+ have good deed in chemiluminescence. It provides a new way to improve the sensitivity through the magnetism of Fe3O4 which can separates and preconcentrates the tested composition.A simple and economical method for sensitive determination of PPi(pyrophosphate) has been established and successfully applied to determine pyrophosphate acid in real sample.We also prepared the nano-particles of Fe3O4 with no surface-modified and detected the molecular of Fe3O4 using the method of chemiluminescence. The limit (3σ,n=11) is 3.0×10-9 mol/L; The Fe3O4 is hard to be used in analysis of biochemical test for the poor water-solubility. So,we prepared amine (carboxyl)-functionalized magnetite nanoparticles With 1,6-hexanediamine (polyacrylic acid) as amine (carboxyl)-functionalization reagent. The amine (carboxyl) not only improves the water-solubility but slao provides the possibility to couple the biological molecules. The functional Fe3O4 can be used as double functions probe (detection, separation and preconcentration) in immunoassay, DNA hybridization and SNP analysis.
