T lymphoma invasion and metastasis,Tiam1 was separated from T lymph cell and expresses Tiam1 protein and is guanine nucleotide exchang factor,GEF,In vivo and vitro,activating Rac1 of Rho GTPase family specifically,going through signaling molecule transduction as JNK,p38MAPK and so on,activating the downstream molecule as POR1,WAVE,PAK1,MLK and so on and plays many kinds of biological function such as regulating remodeling of cytoskeletal structure, influcencing cellular body polarizing process,promoting cellular movement and migrating,participating in regulation of expression and cell proliferation and apoptosis and so on.In nomal tissue,Tiam1 gene only expresses in nomal brain tissues and testiculus tissues highly,but expesses lower or negativly in other normal tissues.In recent years,many researches demonstrates that Tiam1 gene expresses higher in many malignant tumors such as T,B lymphoma,breast cancer,pancreatic cancer,carcinoma of bladder,ovarian cancer,cancer of larynx and lung cancer and so on,the level of expression is relative to invasion and metastasis of tumor cells closely. KISS-1 gene that is anti-metastasis gene was found from melanocytes by microcell mediate technology.The research confirms that B16-BL6,HEK293 and CHO-K1 cell line that was transfected by Hot7T175 was processed by KISS-1 protein and stimulates intracellular density of Ca2+sharply.After KISS-1 protein combined with Hot7T175,which can stimulate PLC through Gq protein- couple,generating two kinds of important second messenger as inositol triphosphate3 and diglyceride,both may promote releasing of cellular calcium ion and activation of PLC.The increasing of concentration of cellular cellular calcium ion can inhibit proliferation of tumor cell, inducing differentiation and apoptosis of tumor cell.Gene transfecting experiment confirmed that colony formation ability of breast cancer cell in scirrho-agar culture is relative to the expression of KISS-1 protein negatively.Many researches confirm that KISS-1 is relative to such tumors as carcinoma of bladder,carcinoma of esophagus,breast cancer,melanoma and cancer of pancreas closely.China has the first morbidity and mortality of eophageal carcinoma in the world, Morbidity and mortality of eophageal carcinoma of Henan province are both the first, but Therefor exploring the mechanism of the invasion and metastasis of esophageal carcinoma has been one of research hot spots.With regard to the expression of Tiam1 and KISS-1 in esophageal carcinoma and its significance among them researchs,to this date domestic and foreign documents havenot reports.In order to discuss the mechanisms of esophageal carcinoma,seach valid methods,which can suppress the infiltration and metastasis of esophageal carcinoma and extend survival time of esophageal carcinoma patients,cut downmortality rate.Our research adopts immunohistochemistry sp method,observing the expressions of Tiam1 and KISS-1 protein in esophageal carcinoma tissues of 60 cases,adjacent non-atypical hyperplasia tissues of 30 cases and normal mucosa tissues of 60 cases,Detecing unitedly illuminates furtherly the mechanisms of infiltration and metastasis of esophageal carcinoma and provides theory foundation for blocking invasion and metastasis and detecting of early stage of esophageal carcinoma.Methods1.Using the technology of immunohistochemistry SP to detect the expression of Tiam1 and KISS-1 in the 60 cases of esophageal squamous cell carcinoma,30 cases of atypical hyperplasia tissues of tumor-adjacent and 60 cases of normal mucosa of esophageal carcinoma respectively.2.Statistical analysis:All the dates were analyzed by SPSS 10.0 statistical package,the count information calculated the positive rate,The comparison of positive rates uses the Chi-square;The relation of two variable are analyzed by the spearman level correlation analysis.The level of significant difference isα=0.05.Results1.Immunohistochemical results indicated:the positive expression rate of Tiam1 protein in normal mucosa of esophagus is 1.67%(1/60);The positive expression rate of Tiam1 protein in atypical non-hyperplasia tissues is 30.00%(9/30);The positive expression rate of Tiam1 protein in the carcinoma tissues is 81.67%(49/60),the positive expression rate of Tiaml protein in the carcinoma tissues is compared with that in the propotional adjacent tissues and normal mucosa tissues from each other, There is stastital significance(P<0.05). 2.Immunohistochemical results indicated:the positive expression rate of KISS-1 protein in normal mucosa of esophagus is93.33%(56/60);The positive expression rate of KISS-1 protein in atypical non-hyperplasia tissues is 63.33%(19/30);The positive expression rate of KISS-1 protein in the carcinoma tissues is 35.00%(21/60), the positive expression rate of KISS-1 protein in the carcinoma tissues is compared with that in the propotional adjacent tissues and normal mucosa tissues from each other,There is stastital significance(P<0.05).3.In the 20 cases of metastatic group,the positive rates of expression of Tiam1 protein in the tumor tissues are 100.00%(26/26);in the non-metastatic group,they are 67.65%(23/34).There is a significant difference between the two groups(P<0.05).4.In the 20 cases of metastatic group,the positive rates of expression of KISS-1 protein in the tumor tissues are 15.38%(4/26);in the non-metastatic group, they are 50.00%(17/34).There is a significant difference between the two groups (P<0.05).5.In the tissues of esophageal squamous cell carcinoma,the positive radio of expression of Tiam1 protein on the low-muscle invaded,deep-muscle invaded and outer lay invaded is 66.67%(10/15),70.00%(14/20) and 100.00%(25/25) respectively.The expression of Tiam1 in the carcinoma tissues with the outer lay invaded is higher than that in the tissue with low-muscle invaded and deep-muscle invaded.There was a significant difference among the groups.(P<0.05).Thus the expression of Tiam1 in the carcinoma tissues with deep-muscle is compared with that with low-muscle,there is no significance(P>0.05).6.In the tissues of esophageal squamous cell carcinoma,the positive radio of expression of KISS-1 protein on the low-muscle invaded,deep-muscle invaded and outer lay invaded is 60%(5/15),50.00%(10/20)and 8.00%(2/25) respectively. The expression of KISS-1 in the carcinoma tissues with low-muscle invaded is higher than that in the tissue with outer lay invaded and deep-muscle invaded.There was a significant difference among the groups.(P<0.05).Thus the expression of KISS-lin the carcinoma tissues with deep-muscle is compared with that with deep-muscle lay, there is no significance(P>0.05).7.The relativity of the expression of Tiam1 and KISS-1 was analysized by statistics,Results showed that there is a positive correlation between them(P<0.01).Conclusions 1.The positive rates of the expression of Tiam1 protein in squamous carcinoma tissues of esophagus,atypical hyperplasia tissues of the tumor-adjacent and normal mucosa tissues were low in turn,The expression of Tiam1 in esophageal squamous cell carcinoma tissues with lymphnode metastasis were higher than that in without lymphnode metastasis,It indicaed that Tiam1 participated in occurrence and development of squamous carcinoma of esophagus,it indicates that there was correlation between the expression of protein and metastasis of esophageal carcinoma.2.The positive rates of the expression of KISS-1 protein in squamous carcinoma tissues of esophagus,atypical hyperplasia tissues of the tumor-adjacent and normal mucosa tissues were high in turn,The expression of KISS-1 in esophageal squamous cell carcinoma tissues with lymphnode metastasis were lower than that in without lymphnode metastasis,It indicaed that KISS-1 participated in occurrence and development of squamous carcinoma of esophagus,it indicates that there was correlation between the expression of protein and metastasis of esophageal carcinoma.3.The positive rates of the expression of Tiam1 protein were different in esophageal squamous tissues with different invasion depth,the positive rates of carcinomas tissues were increased gradually accompany with the invasion depth,it indicated that there was correlation between the expression of Tiam1 protein and invasion of esophageal squamous.4.The positive rates of the expression of KISS-1 protein were different in esophageal squamous tissues with different invasion depth,the positive rates of carcinomas tissues were decreased gradually accompany with the invasion depth,it indicated that there was correlation between the expression of KISS-1 protein and invasion of esophageal squamous.
