Objective The present study was to investigate the role of caveolin-1 on the proliferation and differentiation of osteoblastic MC3T3-E1 .Methods 1.Osteoblastic MC3T3-E1 were cultured in medium involvingÎ'-GP and ascorbic acid.On different growing day (3 ,6 ,9 ,12 ,15 ,18 the day),we observed the expression of caveolin-1 protein via Western Blotting.2.We use the RNAi plasmid pGenesil-1.1-cav-1 to down-regulate the expression of caveolin-1 . The proliferation of osteoblastic MC3T3-E1 was determined using MTT assay after the transfection on third day.The differentiation of osteoblastic MC3T3-E1 was respectively evaluated by ALP activity in cell lysis(PNPP) and osteocalcin(OCN) concentration (ELISA) in medium after the transfection of the aboved two plasmids on 3 ,6 ,9 ,12 ,15 ,18th day.Results 1.The protein expression of caveolin-1 is gradually decreased along with the differentiation of osteoblastic MC3T3-E1.2. There were no significant differences in viability among all groups.The peak value of ALP activity and the concentration of osteocalcin(OCN)in the RNAi group were obviously higher and earlier than control group.however,there were no significant differences in Pcl-neo-cav-1 plasmid group compared with control group.Conclusion Caveolin-1 may down-regulate the differentiation of osteoblastic MC3T3-E1, and has no obvious effect on cell at proliferative phase.
