Changium smyrnioides Wolff is Umbelliferae plant,The subject of studies is the subsidiary subject of foundation research project in Jiangsu technology government agency. Based on the finished research,we use ultrasonic water to extract Changium smyrnioides Wolff.The water extraction liquid is refined by macroporous resin,and isolated and identificated the chemical composition in 30%ethanol which has anti-ADP-induced platelet aggregation in vitro.We separate 5 compounds from this part.They are the D-mannitol,4-Methoxycinnamic acid,(S)2-hydroxyphenylpropionic acid,(R)2-hydroxyphenylpropionic acid,8-methoxy-5-O-β-D-glucosyloxypsoralen;among them,4-Methoxycinnamic acid,(S)2-hydroxyphenylpropionic acid,(R)2-hydroxyphenylpropionic Acid,8-methoxy-5-O-β-D-glucosyloxypsoralen are the first time separated from Changium smyrnioides Wolff.Using SPE to extract the fruit of it,we find 2 compounds.They are Phellopterin(Ⅰ), 5-hydroxyl-8-methoxy- psoralen(Ⅱ),and the first time separated from this plant,too. We use 90%ethanol to extract root bark of Changium smyrnioides Wolff and separate 7 compounds from this part.They are the pentacosanoic acid,Succinic acid,Stigmasterol,Daucosterol,L-Prroglutamic Acid,8-methoxy-5-O-β-D-glucosyloxypsoralen and sucrose.The subject of studies confirme that the water extract of the plant has significantly pharmacological effect on inhibiting platelet aggregation.Application of turbidimetric method to determine each elution spot of it through macroporous resin in vitroanti-ADP -induced platelet aggregation,and the result shows that 30%ethanol macroporous resin elution spot has certain effect on anti-ADP-induced platelet aggergation in vitro finally.We use RP-HPLC external standard quantitative method to mensurate the accumulation content of Phellopterin in different parts of Changium smyrnioides Wolff,we discover the rule that Phellopterin has highest content in the root bark,the root rear part next,lowest in the root without root bark.5-hydroxyl-8-methoxy- psoralen also has higher content in the root bark and fruit than any other parts. The plant is easyly destroyed by insects,so in order to study the difference before and after attack of insects,we adapt the "the Chinese Pharmacopoeia" method to compare the content of water soluble extracts and use HPLC to compare the fingerprints before and after. The result show that water soluble extracts is significantly lower than before,and reduces year by year along with storage time's extension.This study indicates that the quality standards on moth-eaten in current edition of"Chinese Pharmacopoeia" can not carry on the effective monitoring.Changium smyrnioides Wolff and Chuanminshen violaceum quite close in shape,so all previous medical books were recorded mistakably.In this experiment,we compare them through TLC and water soluble extracts,and uses the HPLC to measure the content of Phellopterin that included to both of them,and inspect the component accumulation of different parts to discuss the differences.TLC and water soluble extracts results show significant difference,but the rule of content accumulation rule is similar in a certain extent.This research provides scientific basis to confirm the medicial value,revelate the rule of effective component accumulation,and establish the quality standard of Changium smyrnioides Wolff.
