Houpuwenzhongtang is consisted of Cortex Magnoliae Officinalis, Semen Alpiniae katsumadai, Pericarpium Citri Reticulatae, Radix Aucklandiae, Radix Glycyrrhizae, Poria and Rhizoma Zingiberis. In this paper, the quality assessment, extraction process, pharmcodynamics and pharmacokinetics of the recipe were studied.An HPLC method was developed for the determination of alpinetin and cardamonin in Semen Alpiniae katsumadai. The linearity ranges were 6.0- 120.0μg·mL-1 and 3.0-60.0μg·mL-1 (r≥0.9991), and the recoveries were 99.7 % and 99.8 %, respectively. An HPLC method was developed for the simultaneous determination of six effective components in Houpuwenzhongtang. The linearity ranges were 3.0-60.0μg·mL-1, 7.4-148.0μg·mL-1, 0.75-15.0μg·mL-1, 5.50-110.0μg·mL-1, 7.80-156.0μg·mL-1 and 18.50-370.0μg·mL-1, respectively (r≥0.9991). The recoveries were found to be 99.1 %, 96.6 %, 100.7 %, 96.1 %, 99.2 % and 99.8 %, respectively. HPLC methods were developed for the determination of glycyrrhizic acid and aurantiamarin in Houpuwenzhongtang. The linearity ranges were 25.0-500.0μg·mL-1 (r = 0.9997) and 14.0-280.0μg·mL-1 (r = 0.9995) and the recoveries were 97.0 % and 97.5 %, separately.By investigating the extractive yield of Houpuwenzhongtang and contents of active ingredients such as alpinetin, honokiol, cardamonin, costunolide, dehydrocostus lactone, magnolol, glycyrhizic acid and aurantiamarin, 95 % ethanol was chosen as the best extraction solvent. The optimal alcohol extraction process of Houpuwenzhongtang was studied by orthogonal design with the same inspection index. Three factors were studied in this experiment, including ethanol consumption, duration of extraction and times of extraction. The results showed that the optimal extraction condition was 95 % ethanol consummated 10 times the amount of material, refluxing for twice and 1.5 hours per time.Some pharmacodynamic researches on Houpuwenzhongtang were carried out. The ulcer index and ulcer inhibition ratio were used to evaluate the results in mice and rats, respectively. Compared with the model group, different doses of Houpuwenzhongtang sample were better for ulcer treatment and protection.An HPLC method was developed to determine magnolol in rat plasma with ibuprofen as internal standard. The samples were extracted with acetoacetate. The separation was achieved on a Scienhome C18 (200 mm×4.6 mm, 5μm) column at 30℃. The mobile phase was consisted of methanol -0.5 % glacial acetic acid solution (75:25, v/v) at a flow rate of 1.0 mL·min-1. The UV detection wavelength was set at 225 nm. The mean extraction recoveries exceeded 70.0 %. Linear calibration plot was obtained in the concentration range from 0.08 to 8.00μg 222 mL-1 (r > 0.991). Both intra-day RSD and inter-day RSD were less than 14.0 %. After intragastric administration of Houpuwenzhongtang, magnolol showed to fit two-compartment model,t1/2α, t1/2β,Cmax, AUC0-t were 0.84 h, 13.9 h, 0.52μg·mL-1 and 2.9μg·h·mL-1, respectively.
