The Correlation Of Unscheduled Expression Of Cycline,p27～(kip1) In Multiple Parameters With Clinicpathological Stages In Gastric Carcinoma

Objective: To explore the expression pattern and quantity of cyclinE,p27^kip1 in multiple parameters and the significance with clinicpathologocal stages in gastric carcinoma . At last to reveal the cell cycle principles in gastric carcinoma.Method: There were 34 patients with gastric cacinoma,10 patients with pre-carcinoma lesions and 10 normal gastric mucosae being studied. To make clinical samples into single cell suspensions, mechanical digestion or mixed enzyme digestion of collagenase type I and hyaluronidase was used, then fixed by 80% cold ethanol overnight. After permeated by 0.5% TritonX-100 on ice for 5 minutes,the cell suspensions was added by cyclinE mouse-anti-human monoclonal antibody overnight at 4℃.On the next day ,the cultures was incubated with FITC-conjugated goat-anti-mouse monoclonal antibody to cycinE for 30 minutes , and PE-P27^kip1 Mab for one hour. When DNA dyed by PI and RNA digested by RnaseA in dark room at 20℃for 30 minutes, the single cell suspensions were examined by Flow Cytometry with multiple parameters .Results:1. Aneuploid propotion in normal gastric mucosa,pre-cancer lesions,gastric cancer and para-gastric cancer was 0,20%,76.5% and 17.6% respectively.SPF(S-phase fraction) and PI (proliferation index) in gastric cancer was much higher than in control group(p<0.01).2.The cyclinE and p27^kip1 expression rate in gastric cancer was 32.08%±15.04%,5.58%±4.75%respectively,having considerable difference between that of control group(p<0.01). cyclinE and p27^kip1 expression in gastric cancer was not related with gender,age of patient,Borrmann typing and tumor size, but was intimately correlated with differentiation,depth of invasion,lymphnode metastasis and TNM stage of gastric carcinoma(p<0.05).There was an inverse relationship between cylinE and p27^kip1 expression. The unscheduled expression rate of cylinE in gastric cancer was 52.94%,different from control group.Conclusion: The unscheduled over expression of cyclinE and low or no expression of p27^kip1 during G1/S transition may disturb cell cycle ,and relate to carcinogenesis and progression of gastric carcinoma. Flow cytometry with multiple parameters may disclose the cell cycle principles in gastric carcinoma more completely.
