Objective : To study the expression of membrane typeⅠmatrix metalloproteinase(MMP-14) in squamous cell carcinomas of uterine cervix and to explore their clinicopathological significance.Methods: Tissue microarrays of 24 cases of paired squamous cell carcinoma of uterine cervix,cancer adjacent and adjacent normal tissue and 21 cases of squamous cell carcinoma of cervix,6 adjacent carcinoma tissues and 15 lymph node were investigated immunohistochemically to determine the level of expression of MMP-14 proteins .Results:MMP-14 proteins was noticed to be localized in the cytoplasm and at the cell membrane in carcinoma cell,over expresssion of MMP-14 proteins was found in 41 of 45. Over expression of adjacent carcinoma tissues was found in 3 of 26. Over expression of MMP-14 proteins in lymph node was found in 2 of 25. There was a significant difference between carcinoma and adjacent carcinoma tissues (P=0.000). Expression of MMP-14 protein was significantly increased in lymph node metastasis groups compared with the groups without lymph node metastasis (P<0.025) . Over expression of MMP-14 proteins was associated with size of carcinoma cervicis,invaded muscular deepness and regional lymph node metastasis (P > 0.05) and was not associated with histologic grade,advanced T stage and advanced tumor stage(P>0.05). Conclusion:These results suggest that MMP-14 proteins expression of squamous cell carcinoma of uterine cervix in the cytoplasm and at the cell surface. Over expression of MMP-14 proteins is associated with invasion and metastasis ,and may play a predictive role in lymph node metastasis and early tumor progression.
